Functional characterization of pro-metastatic proteins in breast carcinomas

Abstract:

Tato diplomová práce se zabývá proteinem stathmin 1 (STMN1), TRAF3 interakčním proteinem 2 (TRAF3IP2), ring finger proteinem 25 (RNF25) a PDZ a LIM domény obsahujícím proteinem 2 (PDLIM2). Zmíněné proteiny byly na základě předchozí proteomicko-transkriptomické studie, provedené na klinicko-patologicky reprezentativní soustavě karcinomů prsu, vyhodnoceny jako proteiny potenciálně pro-metastatické. Teoretická část této práce představuje literární rešerši popisující obecný proces vzniku metastáz a také jednotlivé proteiny a jejich buněčné funkce, dále dráhy, do kterých jsou tyto proteiny zapojeny (NF-κB, MAPK/ERK, Wnt a další) a v neposlední řadě jejich vztah k migraci a invazivitě buněk a popis jejich známých interakčních partnerů. Experimentální část práce představují funkční experimenty snažící se objasnit vztah zmíněných proteinů k migraci a invazivitě, dále jejich vztah k epiteliálně – mezenchymální transici a proteomické experimenty zaměřené na detekci jejich interakčních partnerů. Všechny tyto experimenty přispěly k lepšímu pochopení a určení přesnějšího vztahu těchto proteinů k metastazování nádorů.

Abstract:

This diploma thesis is focused on stathmin like protein 1 (STMN1), TRAF3 interacting protein 2 (TRAF3IP2), ring finger protein 25 (RNF25) and PDZ and LIM domain containing protein 2 (PDLIM2). Based on previous proteomics-transcriptomics study on clinico-pathological representative set of breast carcinomas, these proteins were recognized as potentially pro-metastatic. Theoretical part of the thesis summarizes the process of metastasis formation, reviews the functions of mentioned proteins and their involvement in signalling pathways (e.g., NF-κB, MAPK/ERK and Wnt), as well as their known interacting partners. In the experimental part, functional experiments revealing the relationships between these proteins and cell migration, invasiveness and epithelial to mesenchymal transition are described, together with a proteomics-based detection of their interacting partners. All these experiments have contributed to better understanding the potential role of studied proteins in breast cancer metastasis.
