Charakterizace systémů nespecifické transdukce plazmidů u Staphylococcus aureus

Abstract:

Staphylococcus aureus je významným lidským a zvířecím patogenem způsobujícím onemocnění v rozsahu od mírných zánětů kůže až po život ohrožující sepse. Neustále rostoucí počet multirezistentních kmenů tohoto bakteriálního druhu představuje v současnosti závažný celosvětový problém. Rychlá adaptace kmenů S. aureus na používaná antibiotika a antimikrobiální látky je způsobena především horizontálním přenosem genů (HGT) pro rezistenci k těmto látkám. Nejdůležitějším typem HGT u S. aureus je transdukce, při níž je bakteriální DNA přenášena mezi buňkami prostřednictvím temperovaných bakteriofágů. Geny pro rezistenci k antibiotikům se nacházejí často na plazmidech, mobilních genetických elementech (MGE), které se hojně vyskytují napříč různými kmeny S. aureus. Tato práce se věnuje transdukci plazmidů s geny pro rezistenci k antibiotikům u druhu S. aureus. Na základě transdukčních experimentů v klonu USA300 bylo prokázáno, že plazmidy jsou v rámci tohoto klonu přenášeny s vysokou frekvencí, která dosahovala hodnot 10(–5) až 10(–6) CFU/PFU. Klon USA300 je jedním z nejdynamičtěji se vyvíjejících klonů S. aureus a v současnosti je dominantním klonem v USA, kde zahrnuje 97 % všech klinických kmenů S. aureus. Kvantifikací plazmidové DNA přítomné v kapsidách transdukujících bakteriofágů metodou kvantitativní PCR v reálném čase (qPCR) byl stanoven poměr mezi těmito fágy a infekčními fágovými částicemi přibližně 1:1700. Dále byly experimentálně prokázány výborné transdukční schopnosti fága phiJB izolovaného z meticilin rezistentního kmene Jevons B. Byla stanovena kompletní sekvence jeho genomové DNA, která byla následně podrobena analýze in silico. Tímto přístupem bylo identifikováno 70 ORF a byly získány informace o struktuře a funkci většiny jejich produktů. Schopnost fága phiJB přenášet plazmidy byla v sérii transdukčních experimentů porovnána se schopnostmi dalších známých transdukujících bakteriofágů. Bylo zjištěno, že phiJB spolu s fágem phiNM4 z kmene Newman vykazují až o dva řády vyšší frekvence transdukce než ostatní testované fágy. Výsledky práce ukazují, že plazmidy s geny pro rezistenci k antibiotikům se v rámci epidemického klonu S. aureus USA300 mohou pohybovat s vysokou frekvencí, což vytváří podmínky pro vznik a rychlou evoluci multirezistentních kmenů tohoto klonu. Nově popsaný bakteriofág phiJB s výbornými transdukčními schopnostmi by mohl přispět k objasnění molekulárního mechanismu transdukce a k identifikaci genů zapojených do tohoto procesu.

Abstract:

Staphylococcus aureus is a major human and veterinary pathogen causing a variety of diseases ranging from relatively minor skin and soft tissue infections to life threatening sepsis. The ever increasing number of multiresistant strains poses a serious threat to global healthcare system. A fast adaptation rate to antibiotics and antimicrobial compounds in S. aureus is mainly caused by horizontal gene transfer (HGT) of resistance determinants. The most important mechanism of HGT in S. aureus is transduction, in which bacterial DNA is moved from one bacterium to another by temperate bacteriophages. Antibiotic resistance genes are often carried on plasmids, the mobile genetic elements (MGE), that are widely distributed among S. aureus strains. This thesis deals with the transduction of antibiotic resistance plasmids in S. aureus. Based on transduction experiments in the USA300 clone it was proved that plasmids are transferred at high frequencies reaching 10(–5) to 10(–6) CFU/PFU within this clone. The clone USA300 is one of the most expanding and evolving S. aureus clones and nowadays it is a dominant clone in the USA, where it comprises up to 97 % of all clinical S. aureus strains. Quantitative real-time PCR was employed to detect plasmid DNA in transducing phage particles and determine the ratio of transducing particles in phage lysates to infectious phage particles (determined as approximately 1:1700). Further, the outstanding transduction abilities of bacteriophage phiJB isolated from methicillin-resistant S. aureus (MRSA) strain Jevons B were discovered. Sequencing this phage‘s genomic DNA and its detailed in silico analysis revealed 70 ORF as well as the function of most of their products. The ability of phiJB to transfer plasmids was compared to that of other transducing phages. It was found out that phiJB together with phage phiNM4 from the strain Newman can transduce plasmids at up to hundred-fold higher frequencies than other tested bacteriophages. The obtained results show clearly that antibiotic resistance plasmids may spread at high frequency within the epidemic community-associated USA300 clone, which enables emergence and evolution of new multiresistant strains of this clone. The novel bacteriophage phiJB manifesting excellent transduction abilities could contribute to clarifying the molecular mechanism of transduction and to identification of genes involved in this process.
