Preparation of adjuvant nanoliposomal carriers for construction of recombinant vaccines

Abstract:

Tato disertační práce se zabývá přípravou liposomů pro vakcinaci a targeting. Pro vakcinaci byly použity metalochelatační liposomy s navázaným antigenem, pro targeting na CD44 receptor liposomy s navázanou kyselinou hyaluronovou, pro targeting na dendritické buňky byly použity liposomy s manosylovaným povrchem. Pro navázání proteinu na liposomální nosič byla použita metalochelatační vazba mezi lipidem DOGS-NTA-Ni a His-tagem na vybraném proteinu. Pro navázání kyseliny hyaluronové a mananu byl použit nový N-oxy lipid vyvinutý ve spolupráci s RNDr. M. Ledvinou, CSc. (VŠCHT). Skupina RNDr. Ledviny, CSc. připravila také řadu adjuvans vycházejících ze struktury MDP, jejichž imunostimulační účinky byly dále testovány in vivo. Struktura liposomů byla studována pomoci dynamického rozptylu světla, nanoparticle tracking analýzy, TRPS metody, transmisní elektronové mikroskopie, mikroskopie atomárních sil, cryoelektronové mikroskopie, gelové permeační chromatografie, fluorimetrie. Pro studium targetingu byla použita konfokální mikroskopie, fluorescenční mikroskopie, western blotting, průtoková cytometrie. Příprava metalochelatačních liposomů vedla k vývoji experimentální vakcíny, liposomy s kyselinou hyaluronovou úspěšně targetovaly buňky s exprimovaným CD44 receptorem. Manosylované liposomy byly schopny v modelech in vitro navodit stimulaci dendritických buněk, která byla srovnatelná s lipopolysacharidem. Byly vyvinuty nové liposomální platformy pro targeting a vakcinaci, včetně liposomů pro mukoadhezivní aplikace. Teoretická část obsahuje stručný úvod o liposomech, jejich přípravě, možnostech analýzy a jejich praktických aplikacích, se zaměřením na vazbu antigenu na povrch liposomu a využití liposomů k přípravě vakcín. Teoretická část dále obsahuje stručný úvod do tématu biokonjugací, se zaměřením na nový koncept Click chemie. V praktické části je okomentováno 7 článků zabývajících se tématem disertační práce. Těchto 7 článků je přiloženo, v nich jsou do hloubky popsány a diskutovány použité metody a výsledky.

Abstract:

This Ph. D. Thesis deals with preparation of liposomes for vaccination and targeting. Metallochelatig liposomes with bound antigen were used for vaccination, liposomes with bound hyaluronic acid were used for CD44 targeting and mannosylated liposomes were used for targeting dendritic cells. Metallochelating linkage between DOGS-NTA-Ni lipid and His-tag at the end of the selected protein was used for binding of the protein to the liposomal carrier. Novel N-oxy lipid developed in cooperation with RNDr. Miroslav Ledvina, CSc. was used for binding of hyaluronic acid to liposomes, and the same lipid was used for binding mannan. The scientific group of RNDr. Ledvina, CSc. also prepared a series of adjuvants based on the MDP structure. The structure of liposomes was studied using dynamic laser scattering, nanoparticle tracking analysis, tunable resistive pulse sensing method, transmission electron microscopy, atomic force microscopy, cryoelectronic microscopy, gel permeation chromatography and spectrofluorimetry. For targeting studies, confocal microscopy, fluorescent microscopy, western blotting and flow cytometry were used. Preparation of metallochelating liposomes led to the development of an experimental vaccine, liposomes with hyaluronic acid successfully targeted cells with expressed CD44 receptor, mannosylated liposomes were able to induce immunostimulation of dendritic cell in vivo, comparable to lipopolysaccharide. Novel liposomal platforms for targeting and vaccination were developed, including liposomes for mucoadhesive applications. The theoretical part contains a brief introduction about liposomes, their preparation, analysis options and their practical applications, focusing on antigen binding to liposomal surface and using of liposomes to prepare vaccines. The theoretical part further contains a brief introduction to bioconjugation, with a focus on the new concept Click chemistry. In the Practical part, 7 manuscripts dealing with the topic of the dissertation are commented. These 7 manuscripts are attached, the methods used and the results are described and discussed in depth.
