Izolace a kultivace bovinních dendritických buně

Abstract:

Dendritické buňky jsou buňky imunitního systému, které pocházejí z kostní dřeně a mají schopnost prezentovat antigeny naivním T buňkách. Dendritické buňky zajišťují primární imunitní odpověď a nacházejí se jak v lymfoidních, tak i většinou nelymfoidních tkáních. Studium těchto buněk představuje důležité spojení mezi specifickou a nespecifickou částí imunitního systému. Správná aktivace dendritických buněk určuje, jakým způsobem bude patogen eliminován. Následující disertační práce se zaměřuje na vývoj dendritických buněk a metody jejich izolace z krve skotu. Práce je rozdělena do tří experimentů. První experiment porovnává dvě metody izolace monocytů, které slouží pro vývoj dendritických buněk. Druhý experiment zkoumá možnost vývoje a kultivace dendritických buněk na polykaprolaktonových vláknech. Třetí experiment se zaměřuje na izolaci myeloidních dendritických buněk přímo z krve a jejich detekci viru bovinní leukémie. Výsledky této disertační práce přispějí k lepšímu porozumění izolace a kultivace dendritických buněk a mohou mít budoucí potenciál v oblasti imunologie

Abstract:

Dendritic cells are cells of the immune system that originate in the bone marrow and have the ability to present antigens to naïve T cells. Dendritic cells provide the primary immune response and are found in both lymphoid and mostly non-lymphoid tissues. The study of these cells represents an important link between the specific and non-specific parts of the immune system. Proper activation of dendritic cells determines how the pathogen will be eliminated. The following dissertation focuses on the development of dendritic cells and methods of their isolation from bovine blood. The work is divided into three experiments. The first experiment compares two methods of isolation of monocytes, which are used for the development of dendritic cells. The second experiment investigates the possibility of developing and cultivating dendritic cells on polycaprolactone fibres. The third experiment focuses on the isolation of myeloid dendritic cells directly from blood and their detection of the bovine leukaemia virus. The results of this dissertation will contribute to a better understanding of the isolation and culture of dendritic cells and may have future potential in the field of immunology
