The role of hormones in tumor development, the role of androgen receptors in AGR2 signaling in breast cancer.

Abstract:

Předložená práce se na teoretické úrovni zabývá úlohou hormonů ve vzniku a vývoji nádorových onemocnění se zaměřením na karcinom mléčné žlázy, vaječníků, endometria, prostaty, nádory varlat, štítné žlázy a osteosarkom. Část práce je věnována popisu a vlastnostem potenciálního nádorového biomarkeru Anterior gradient 2 (AGR2), především s důrazem na mechanismy regulace jeho exprese v nádorové buňce. Mechanismy regulace exprese AGR2 nebyly doposud zcela objasněny, ale je známo, že v některých hormon-dependentních nádorech je exprese AGR2 řízena prostřednictvím hormonů, konkrétně prostřednictvím estrogenů u karcinomu mléčné žlázy a androgenů u karcinomu prostaty. Cílem praktické části bylo studium nových mechanismů, které se podílejí na regulaci exprese AGR2 u karcinomu mléčné žlázy se zaměřením na úlohu androgenního receptoru. V první části experimentální práce byla stanovena exprese proteinu AGR2 a androgenních receptorů ve vybraných buněčných liniích odvozených od karcinomu mléčné žlázy metodami imunofluorescenčního barvení in situ a PAGE elektroforézy s Westernovým přenosem. V druhé části byl studován vliv 5α-dihydrotestosteronu na hladinu proteinu AGR2 v buněčných liniích MCF-7 a MDA-MB-453, obou odvozených od karcinomu mléčné žlázy. Dosažené výsledky potvrdily přítomnost proteinu AGR2 a androgenních receptorů v buněčných liniích MCF-7, T-47D a MDA-MB-453, přičemž bylo zjištěno, že aktivita androgenních receptorů zřejmě neovlivňuje hladinu sledovaného proteinu v karcinomu mléčné žlázy.

Abstract:

The theoretical part of the thesis describes the role of hormones in carcinogenesis with the focus on breast, ovarian, endometrial, prostate, testicular, thyroid cancers and osteosarcoma. The attention is paid also to the description and the properties of potential cancer biomarker Anterior gradient 2 (AGR2), especially to the mechanisms that are responsible for AGR2 expression regulation in tumor cell. Although those mechanisms have not been fully explored yet, there are some studies proving a role of hormones in regulation of AGR2 expression in some hormone-dependent cancers, namely the role of estrogens in breast cancer and androgens in prostate cancer. The aim of the thesis is to study new mechanisms of AGR2 expression regulation in breast cancer, especially the role of androgen receptor. The first part of the experimental work includes an assessment of AGR2 and androgen receptor expression in particular breast cancer cell lines by the methods of immunofluorescence staining in situ and PAGE electrophoresis combined with Western blot. In the second part the influence of 5α-dihydrotestosterone treatment on the AGR2 expression in cell lines MCF-7 and MDA-MB-453 derived from breast cancers is studied. According to our results, AGR2 and androgen receptor expression was proved in MCF-7, T-47D and MDA-MB-453 cell lines however it seems that AGR2 expression is independ on activation of androgen receptors in breast cancer.
