Expression of structural proteins of animal viruses in the eucaryotic systems

Abstract:

Abstrakt disertační práce V eukaryotním systému exprese heterologních proteinů byly zkonstruovány rekombinantní bakuloviry exprimující kompletní gen ORF7 evropského a severoamerického genotypu viru reprodukčního a respiračního syndromu prasat (PRRSV), který kóduje strukturální nukleokapsidový protein N. Získané rekombinantní proteiny byly použity jako antigeny k sestavení nepřímého ELISA testu (nELISA) k průkazu PRRSV protilátek v sérech prasat. Paralelní vyšetření souboru 552 sér prasat nELISA testem a standardní diagnostickou soupravou HerdChek PRRS ELISA stanovilo 96,3 % citlivost a 92,6 % specifičnost nELISA testu. Aplikace tohoto testu byla potvrzena také při sérologické depistáži v inseminačních stanicích kanců a v karanténách v ČR. Rozsah genetické variability v oblasti genů ORF6-7 byl sledován u 3 modifikovaných živých vakcín, 3 PRRSV izolátů a 6 terénních virů zachycených v České republice a na Slovensku technikou délkového polymorfismu restrikčních fragmentů (RFLP). Set restrikčních enzymů Hae II, Alu I a BsaJ I umožnil spolehlivě rozlišit viry evropského a severoamerického genotypu, ale i viry shodného genotypu. Nejvíce častým štěpícím RFLP vzorem byl kód 1-1-1 mezi viry evropského genotypu a odpovídal všem terénním virům, izolátu CAPM V-502 a vakcinačnímu kmeni Porcilis PRRS. Výsledky této analýzy mohou být využity při rozhodování o výběru vakcinačního kmene s maximální genetickou podobností k cirkulujícím terénním virům PRRS. V rámci vývoje a standardizace techniky kvantitativní RT-PCR pro detekci a charakteristiku viru PRRS byl připraven specifický RNA transkript genu ORF6. Jeho konstrukce byla nezbytná k sestavení kalibrační přímky pro odečet absolutní kvantifikace virových molekul RNA referenčního kmene Lelystad a 5 českých terénních izolátů PRRSV. Opakovanou TaqMan RTPCR analýzou ředěných RNA transkriptů bylo detekováno 5-50 kopií genu ORF6 kódujícího strukturální protein M. Tato vysoce specifická a citlivá technika může být použita jako diagnostický nástroj pro kvantifikaci geneticky příbuzných kmenů viru PRRS. Metoda kvantitativní RT-PCR se značením SYBR Green I byla ověřena pro genetickou identifikaci RNA templátů a kvantifikaci cDNA virů bovinní virové diarrhoey (BVDV) v infikovaných buněčných kulturách a v klinických vzorcích perzistentně infikovaného skotu. Hladiny RNA/BVDV byly spolehlivě monitorovány nejen v bazénových vzorcích mléka a v sérech perzistentně infikovaného skotu, ale i v šarži telecího séra určeného ke kultivaci buněčných kultur. Získané výsledky potvrdily metodu kvantitativní RT-PCR jako vysoce citlivou pro kvantifikaci a stanovení stupně variability BVDV.

Abstract:

Dissertation Abstract In the eucaryotic system of heterologous protein expression, recombinant baculoviruses expressing complete ORF7 gene of European and North American genotype of porcine reproductive and respiratory syndrome virus (PRRSV) that encodes structural nucleocapsid protein N were constructed. The obtained recombinant proteins were used as antigens in the developed indirect ELISA test (nELISA) for detection of PRRSV antibodies in porcine serum. Simultaneous examination of a total of 552 pig serum samples by the nELISA test and the standard diagnostic kit HerdChek PRRS ELISA revealed 96.3% sensitivity and 92.6% specificity of the nELISA test. Applicability of this test was also confirmed during serological screening in the artificial insemination boar stations and the quarantined animals in the Czech Republic. The range of genetic variability of ORF6-7 genes was investigated in 3 modified live vaccines, 3 PRRSV isolates and 6 field viruses detected in the Czech Republic and in Slovakia by the restriction fragment length polymorphism (RFLP) technique. The set of restriction enzymes Hae II, Alu I and BsaJ I allowed reliable differentiating between viruses of European and North American genotypes and between viruses of identical genotype. The RFLP code 1-1-1 was the most frequent digestion pattern in viruses of the European genotype and was identical with all field viruses, the isolate CAPM V-502 and the vaccine strain Porcilis PRRS. The results of this analysis can be used in selecting a vaccine strain with maximum genetic similarity to the circulating field PRRS viruses. During development and standardisation of the quantitative RT-PCR technique for detection and characterisation of PRRS virus, specific RNA transcript of ORF6 gene was prepared. Its construction was necessary to make the calibration curve for reading absolute values of viral RNA molecules of the reference strain Lelystad and 5 Czech field isolates of PRRSV. By the repeated TaqMan RT-PCR analysis of the diluted RNA transcripts, 5-50 copies of ORF6 gene encoding the structural protein M were detected. This highly specific and sensitive technique can be used as a diagnostic tool for quantification of genetically related strains of PRRS virus. The quantitative RT-PCR method with SYBR Green I labelling was tested for genetic identification of RNA templates and quantification of cDNA viruses of bovine viral diarrhoea (BVDV) in the infected cell cultures and in clinical samples from persistently infected cattle. The RNA/BVDV levels were reliably monitored not only in bulk milk samples and sera from persistently infected cattle, but also in a batch of calf serum intended for cell culture. The obtained results confirmed the quantitative RT-PCR method to be highly sensitive for quantification and assessment of the degree of variability amongst BVDV.
