Determination of functional activity of lymphocytes using tests detect interferon gamma

Abstract:

Testy založené na detekci interferonu-gama se dnes běžně používají v laboratořích pro diagnostiku některých infekčních onemocnění. Řadí se mezi testy nepřímé diagnostiky, průkaz reakce buněčné imunity na přítomnost specifického antigenu testovaného infekčního agens, a proto by diagnostika neměla záviset pouze na jejich výsledku. Principem tohoto typu testů je detekce interferonu gama produkovaného T-lymfocyty senzibilizovanými specifickými antigeny pro testované infekční agens. V této práci je popsán princip IGRA testů včetně jejich dvou modifikací a možností uplatnění při diagnostice infekčního agens. Praktická část popisuje provedení testů pro infekční onemocnění tuberkulózu. Druhá část se zabývá srovnáním výhod a nevýhod obou modifikací testů QuantiFERON a T-SPOT, ze kterých vychází doporučení, kterou metodu zvolit v konkrétním případě.

Abstract:

Interferon-gamma detection assays are nowadays commonly used in laboratories to diagnose some infectious diseases. They are line up among tests of indirect diagnostics, the demonstration of the response of cellular immunity to the presence of a specific antigen of the tested infection, and therefore the diagnosis should not depend only on their result. The principle of this type of assay is the detection of interferon gamma produced by T-lymphocytes sensitized by specific antigens for the infectious agent tested. This work describes the principle of IGRA tests, including their two modifications and the possibility of application in the diagnosis of infectious agents. The practical part describes the performance of tests for infectious tuberculosis. The second part deals with a comparison of advantages and disadvantages of both modifications of the QuantiFERON and T-SPOT tests, on which the recommendation of which method to choose in a particular case is based.
