Microorganisms on dust particles and their effects on human airway microbiome

Abstract:

Prachové částice představují významný rezervoár mikroorganismů, kterému je člověk denně vystavován. Vdechnutím těchto mikroorganismů může dojít ke kolonizaci respiračního traktu a následnému rozvoji onemocnění. Mikrobiom prachu je v různých typech budov odlišný. Ve zdravotnických zařízeních jsou častěji využívány dezinfekční prostředky než v kancelářských budovách nebo domácnostech, což může mít významný vliv na složení mikroorganismů v prachu. Za účelem popisu složení prachu z různých prostředí a lepšího porozumění vlivu mikrobiomu prachu na mikrobiom dýchacích cest byly v rámci diplomové práce odebrány vzorky prachu z domácností a pracoviště účastníků studie a jejich stěry z nosohltanu. Diplomová práce se zaměřuje na optimalizaci postupů a zpracování vzorků prachu a stěrů z nosohltanu. Vzorky byly po izolaci DNA a amplifikaci genu pro 16S rRNA osekvenovány na přístroji MiSeq (Illumina) a získaná data analyzována. Výsledky analýzy poskytly informaci o tom, do jaké míry je mikrobiom dýchacích cest ovlivněn prostředím, ve kterém lidé tráví většinu času.

Abstract:

Dust particles represent significant reservoir of microorganisms which humans are exposed to every day. By inhalation of these microorganisms, colonization of the respiratory tract and subsequent development of the disease may occur. Dust microbiome differs in different types of buildings. Disinfectants are used more frequently in healthcare facilities than in office buildings or houses, that can have a significant impact on the composition of microorganisms in dust. In order to describe the composition of dust from different environments and better understanding the effect of dust microbiome on the microbiome of respiratory tract, samples of dust from households and workplace and nasopharyngeal swabs were taken from volunteers as a part of this study. The diploma thesis focuses on the optimization of procedures and processing of dust samples and nasopharyngeal swabs. Samples were sequenced on a MiSeq device (Illumina) after DNA isolation and 16S rRNA gene amplification, the obtained data were analysed. The results of the analysis provided information on the extent to which the airway microbiome is affected by the environment where people spend most of their time.
