Characterization of putative interaction partners of telomerase in Arabidopsis thaliana model plant

Abstract:

Telomerase is an enzyme with a reverse transcriptase function, that compensates for the loss of telomeric DNA that occurs during chromosome replication, but it is supposed to act also outside the telomeres. A previous study identified a new telomerase interaction partner – ARM protein containing armadillo/β-catenin-like repeats. In this study, we investigated the effect of the position of T-DNA insertion in the ARM gene on telomere length and the level of ARM transcript. It was shown that the defect in the ARM function did not affect the length of telomeres when compared to the wild type plants of the Columbia ecotype. We further examined the effect of DNA damaging agents on early plant development, telomerase activity and the transcription of selected genes involved in homologous recombination in A. thaliana mutant lines with T-DNA insertion in the second intron of the ARM gene. Phenotypic analysis of the root length showed that seedlings of arm mutant tolerated better the damage caused by the hydroxyurea. Telomerase activity was comparable in all samples, which strengthened the hypothesis of the role of ARM in the non telomeric functions of telomerase. Analysis of the relative levels of transcripts of genes involved in homologous recombination revealed the possible function of ARM protein as a negative regulator of transcription of these genes. This study is a part of a broader research project focused on putative non-telomeric functions of plant telomerase.

Abstract:

Telomeráza je enzým s funkciou reverznej transkriptázy, ktorý nahrádza straty telomérovej DNA vznikajúce pri replikácii chromozómov, ale pôsobí aj mimo telomér. V predchádzajúcej štúdii bol identifikovaný nový interakčný partner telomerázy – proteín ARM obsahujúci vo svojej štruktúre „armadillo/β-catenin-like“ repetície. V tejto práci sme skúmali vplyv polohy T-DNA inzercie v géne kódujúcom proteín ARM na dĺžku telomér a na hladinu transkriptu tohoto génu. Zistili sme, že defekt vo funkcii ARM neovplyvňuje dĺžku telomér v mutantných rastlinách v porovnaní s rastlinami štandardného typu, ekotypu Columbia. Ďalej sme sa zaoberali vplyvom látok spôsobujúcich poškodenie DNA na dĺžku korienkov v rannom vývojovom štádiu rastlín, aktivitu telomerázy a transkripciu vybraných génov zapojených do procesov homologickej rekombinácie v mutantných líniách A. thaliana s T DNA inzerciou v 2. intróne génu ARM. Fenotypová analýza dĺžky korienkov ukázala, že klíčne rastliny arm mutantov lepšie znášali poškodenie spôsobené hydroxymočovinou. Aktivita telomerázy bola porovnateľná vo všetkých vzorkách, čo posilnilo predpoklad úlohy ARM pri netelomérových funkciách telomerázy. Analýza relatívnych hladín transkriptov génov súvisiacich s homologickou rekombináciou odhalila, že proteín ARM funguje pravdepodobne ako negatívny regulátor expresie týchto génov. Táto štúdia je súčasťou širšieho výskumu zaoberajúceho sa prípadnými netelomérovými funkciami rastlinnej telomerázy.
