Genetic association case-control studies - gastroesophageal reflux disease

Abstract:

Tato diplomová práce se věnuje genetickému pozadí multifaktoriální gastroezofageální refluxní choroby, jejíž incidence v západních zemích neustále stoupá. Pro asociační studie kontrol a případů byly na základě literární rešerše vybrány geny kódující epidermální růstový faktor (EGF) a jeho receptor (EGFR), receptor možné vaniloidové podrodiny členu 1 (TRPV1) a pohlavní hormony vázající globulin (SHBG) a konkrétní jednonukleotidové polymorfizmy (SNPs) v těchto genech, které by se mohly zapojit do etiopatogeneze tohoto onemocnění. Dále byly zjišťovány plazmatické koncentrace celkového testosteronu a estrodiolu, volného testosteronu, SHBG a EGF. V této studii byla alela A SNP +61 EGF A>G (rs4444903) spojena s rizikem refluxní ezofagitidy. Dále byla nižší plazmatická koncentrace celkového i volného testosteronu asociována s vyšším rizikem adenokarcinomu jícnu u mužů.

Abstract:

This diploma thesis deals with the genetic background of multifactorial gastroesophageal reflux disease, whose incidence is raising in Western countries. For case-control association studies, the genes coding epidermal growth factor (EGF) and its receptor (EGFR), transient receptor potential vanilloid subtype 1 (TRPV1) and sex hormone-binding globulin (SHBG) and its specific single nucleotide polymorphisms (SNPs) that could be involved in the etiopathogenesis of this disease, were selected based on the literature search. Plasma concentrations of total testosterone and estrodiol, free testosterone, SHBG and EGF were also determined. In this study, allele A of SNP +61 EGF A>G (rs4444903) was associated with risk of reflux esophagitis. Furthermore, lower plasma concentrations of total and free testosterone were connected with a higher risk of esophageal adenocarcinoma in men.
