Archaea and their potential role in human disease

Abstract:

Doména Archaea byla od svého objevení po dlouhou dobu považována za různorodý soubor kolonizátorů extrémního prostředí, jejichž vliv byl pro lidské zdraví zanedbatelný. Až v poslední době, zejména s rozvojem citlivějších technik a porozumění problematice kultivace těchto mikroorganizmů se ukázalo, že jsou nejen součástí přirozeného lidského mikrobiomu, ale mohou mít i vliv na lidské zdraví. Řada publikací experimentálně prokázala schopnost archaeí interagovat s lidským imunitním systémem a vyvolat jeho odpověď, přičemž došlo k popsání antigenních struktur i samotných mechanizmů. Dokázán byl i přímý vztah mezi přítomností archaea a závažností infekcí ústní dutiny, stejně jako spojitost mezi jejich výskytem a diverzitou při onemocněních gastrointestinálního traktu. Průkaz archaea přímo v infekčních ložiscích může rovněž napovídat, že jsou tyto mikroorganizmy nějakým způsobem zapojeny do mechanizmů lidské patogeneze. Tato práce shrnuje dosavadní poznatky o problematice výskytu archaea a jejich případném vztahu s lidskými onemocněními.

Abstract:

Since its discovery, the domain Archaea has been recognized as a diverse group of colonizators of the extremophilic environment, which impact on human health was negligible. Only recently, with improvement in detection sensitivity and a better understanding of specific cultivation techniques, it turned out that archaea create a substantial part of the human microbiome and may impact the human health. The recent publications have shown, that archaea are capable of specific interactions with the human immune system and may provoke an immune response. Regarding this, the antigenic structure, as well as mechanism behind the process was identified. A direct relationship between the population of archaea and the severity of oral infection has been proven, as well as a possible link between archaeal abundance and several gastrointestinal disorders. Evidence of archaea as a part of human infectious consortia also support the hypothesis, that the archaea may be a part of pathogenic mechanisms. Thus this work aims to summarize current knowledge concerning the possible relationship between archaea and human pathogenesis.
