The Negative Acute Phase Reactants of Inflammation

Abstract:

Teoretická část bakalářské práce se zabývá zánětem, reaktanty akutní fáze zánětu a železem. Kapitola reaktanty akutní fáze se nejvíce věnuje negativním markerům zánětu, a to prealbuminu, albuminu a nejvíce transferinu. Nejen u těchto, ale i u ostatních markerů zánětu jsou popsány jejich vlastnosti, funkce, diagnostika a metody stanovení. Praktická část práce se zaměřuje na srovnání dvou výpočetních metod saturace transferinu. Pro výpočet saturace se musely změřit koncentrace transferinu, železa a volné vazebné kapacity. Změřilo se 98 náhodně vybraných vzorků z Ústavu klinické biochemie ve Fakultní nemocnici v Ostravě-Porubě. Stanovení transferinu se provádělo na imunochemickém analyzátoru Immage. Železo a volná vazebná kapacita se stanovovalo na analyzátoru AU5400. Vypočítané hodnoty byly statisticky zpracovány a vyhodnoceny. Ze statistického vyhodnocení vyplynulo, že rozdíly mezi výpočetními metodami jsou statisticky významné. Z klinického hlediska jsou však tyto rozdíly zanedbatelné.

Abstract:

Theoretical part of bachelor thesis is about inflammation, acute phase reactants of inflammation and iron. Chapter called Acute Phase Reactants describes the most devoted negative markers of inflammation prealbumin, albumin and, especially, transferrin. Also other markers of inflammation and their features, functions, methods of diagnosis and determination are explained in this chapter. The practical part is focused on the comparison of two methods used for computing transferrin saturation. To calculate the saturation, the concentration of transferrin, iron and unsaturated iron-binding capacity had to be measured. 98 randomly selected samples from the Department of Clinical Biochemistry at the University Hospital in Ostrava-Poruba has been measured. Determination of transferrin was carried out on immunochemical analyzer IMMAGE. Iron and unsaturated iron-binding capacity were then carried out by using the analyzer AU5400. The calculated values were statistically processed and evaluated. Statistical evaluation showed that differences among the computational methods are statistically significant. From a clinical perspective, these differences are insignificant.
