The Potential Of 3D Printing For Supression Of Alcohol-induced Dose Dumping Effect Of Matrix Tablets Containing Tramadol Hydrochloride

Abstract:

Tato diplomová práce se zabývá potenciálem 3D tisku pro potlačení alcohol-induced dose dumping efektu matricových tablet s tramadol hydrochloridem, jakožto modelovou účinnou látkou. Matricové tablety byly připraveny metodou přímého lisování. Jako retardující komponenta v tabletách byl použit Compritol? 888 ATO a jako lubrikant byl použit stearát hořečnatý. Pro potlačení alcohol-induced dose dumping efektu byly 3 formulace tablet potaženy pomocí metody 3D tisku. Pro potažení tablet byl použit celulózový polymer AffinisolTM HPMC HME 15 LV. Disoluční chování potahovaných tablet v disolučním médiu s obsahem 40 obj. % alkoholu bylo srovnáno s profilem nepotahovaných tablet. Disoluční test byl proveden dle Českého lékopisu. Probíhal při teplotě lidského těla, tj. 37 °C, první dvě hodiny v médiu s obsahem alkoholu (40 obj. %, pH 1,2), poté byly tablety umístěny do disolučního média o pH = 1,2, simulující žaludek, již bez přítomnosti alkoholu. Uvolněné množství tramadol hydrochloridu bylo stanovováno pomocí UV/VIS spektrometrie a získaná data byla kvantitativně vyhodnocena pomocí vhodných matematických modelů. Na základně získaných kinetických parametrů byl vyhodnocen účinek potahu tablety a potlačení alcohol-induced dose dumping efektu.

Abstract:

This master's thesis is focused on the potential of 3D printing for suppression of alcohol-induced dose dumping effect of matrix tablets containing tramadol hydrochloride as a model active substance. Matrix tablets were prepared by direct compression. Compritol? 888 ATO was used as a retarding component in tablets and magnesium stearate was used as a lubricant. 3 tablet formulations were coated by the method of 3D printing to suppress alcohol-induced dose dumping effect. Cellulose polymer AffinisolTM HPMC HME 15 LV was used to coat the tablets. Dissolution behaviour of coated tablets in dissolution medium containing 40 vol. % of alcohol was compared with the profile of uncoated tablets. Dissolution test was realized according to the Czech Pharmacopoeia. The test was carried out at human body temperature, i.e. 37 °C, for the first two hours in an alcohol-containing medium (40 vol. %, pH 1.2), then the tablets were placed in a dissolution medium of pH = 1.2, simulating a stomach which was already without presence of alcohol. Released amount of tramadol hydrochloride was determined by UV/VIS spectrometry and obtained data were quantitatively evaluated using suitable mathematical models. Based on the obtained kinetic parameters, the effect of tablet coating and the suppression alcohol-induced dose dumping effect were evaluated.
