Příprava prekurzorů antikoagulancia Rivaroxabanu s využitím asymetrické Henryho reakce

Abstract:

Tato diplomová práce se zabývá syntézou orálního antikoagulancia (S)-Rivaroxabanu (1), která je založená na aplikaci asymetrické Henryho reakce. Hlavním cílem práce bylo především studovat asymetrickou Henryho reakci na dvou rozdílně substituovaných substrátech (aldehydech) 81 a 93 při použití různých enantioselektivních katalyzátorů. Výsledkem této studie měla být maximalizace enantiomerní čistoty nitroaldolových intermediátů 82 a 94 a následně finálního léčiva 1. Do studie bylo zařazeno celkem pět účinných enantioselektivních katalyzátorů na bázi měďnatých komplexů chirálních ligandů 84a-86a, 89 a 96. Za zvolených reakčních podmínek bylo dosaženo vysokých enantiomerních přebytků (83-91 % ee) v nitroaldolech 82 a 94. Bylo zjištěno, že enantioselektivita dosažená s jednotlivými katalyzátory se pro jednotlivé aldehydy 81 a 93 neliší. Dále byla ověřena možnost separace epimerů v případě O-bornylových derivátů 94 a 95, a to pomocí chromatografie nebo krystalizace. Bohužel, vhodné podmínky pro separaci epimerů meziproduktů léčiva Rivaroxaban 94 resp. 95 se v rámci této diplomové práce nepodařilo naleznout.

Abstract:

This thesis deals with the synthesis of oral anticoagulant drug (S)-Rivaroxaban (1) based on the application of asymmetric Henry reaction. The main aim of this work was the study of Henry reaction using two differently substituted substrates (aldehydes) 81 and 93, in which the various enantioselective catalysts were utilized. The result of this study should lead to maximizing of enantiomeric purity of nitroaldol intermediates 82 and 94 as well as the final drug 1. The study included the five highly efficient enantioselective catalysts based on copper(II) complexes of chiral ligands 84a-86a, 89 and 96. High enantiomeric excess (83-91 % ee) in the nitroaldols 82 and 94 were achieved under used reaction conditions. It was found, that the enantioselectivity obtained with the individual catalysts in application of both aldehydes 81 and 93 is similar. Further, the separation of epimers of O-bornyl derivatives 94 and 95 were tested by means of chromatography and crystallization. Unfortunately, the conditions suitable for separation of epimers of intermediates of drug Rivaroxaban 94 and 95 were not found.
