Mutagenesis of RSL lectin from phytopathogen Ralstonia solanacearum

Abstract:

Tato práce byla zaměřena na studium vlivu aromatické aminokyseliny na vazbu ligandu s proteinem. Tento vliv byl studován pomocí mutantních forem proteinu se záměnami aminokyselin na patřičných místech. Mutanti byli připraveni pomocí místně cílené mutageneze in vitro, což je metoda široce používaná pro produkci proteinů nesoucích požadované vlastnosti. Lektin RSL, který byl vybrán pro tuto práci, pochází z fytopatogenu Ralstonia solanacearum napadajícího převážně zemědělské rostliny. Tento fukosa specifický lektin sloužil jako modelový protein pro sledování vlivu aromatické aminokyseliny tryptofanu na interakci lektinu se sacharidovým ligandem. Postupně byla provedena substituce této aminokyseliny nejdříve v jednom a poté i v druhém vazebném místě monomerní formy lektinu. Třetí typ mutanta byl protein nesoucí záměnu v obou vazebných místech najednou. Vliv mutací na vazebnou afinitu a specifitu připravených mutantů byl testován pomocí metod ITC (izotermální titrační mikrokalorimetrie) a SPR (rezonance povrchového plasmonu). Také byla prováděna krystalizace připravených mutantů.

Abstract:

This thesis was focused on study of an aromatic amino acid effect on protein – ligand interactions. This effect was studied using mutant proteins with amino acid substitution in desired positions. The mutant proteins were created by site-directed mutagenesis in vitro, an important technique widely used for producing proteins with improved or altered properties. The plant pathogen Ralstonia solanacearum causes lethal wilt mostly in agricultural plants. This bacterium produces L-fucose-binding lectin RSL, which was used for understanding of tryptophan influence on sugar – lectin interactions. First tryptophan was substituted in one binding site, second in the next binding site. Third type of the mutant protein was a double mutant. ITC (isothermal titration microcalorimetry) and SPR (surface plasmon resonance) were utilized for testing effects of mutation on binding affinity and specificity of mutated proteins. Mutant proteins were also crystallized.
