Characterization of NK cells in patients with endometriosis

Abstract:

Endometrióza je progresivní chronické onemocnění, pro které je charakteristický výskyt funkční endometriální tkáně mimo dutinu děložní. Jedná se o velmi časté onemocnění, které postihuje až 15 % žen v reprodukčním věku, u kterých vede nejčastěji k vzniku chronické pánevní bolesti, ale i neplodnosti. Patogeneze endometriózy není stále zcela známa, existuje ale několik teorií, které se snaží její vznik osvětlit. Jednou z nich je i přítomná dysregulace imunitního systému. V této bakalářské práci jsme pomocí průtokové cytometrie porovnávali zastoupení NK buněk v periferní krvi a jejich povrchových KIR receptorů u žen trpících endometriózou a zdravých žen. Bohužel i přes důkladnou analýzu jsme nedošli k žádnému statisticky významnému rozdílu mezi sledovanými soubory. Je třeba provést další studie s větším množstvím vzorků, které by mohly lépe zachytit biologickou variabilitu a použít komplexnější metodologii analýzy.

Abstract:

Endometriosis is a progressive chronic condition characterized by the presence of functional endometrial tissue outside the uterine cavity. It is a very common disorder, affecting up to 15 % of women of reproductive age, often leading to chronic pelvic pain and infertility. The pathogenesis of endometriosis is not fully understood, although several theories attempt to elucidate its origins. One of them involves the dysregulation of the immune system. In this bachelor thesis, we compared the representation of NK cells in peripheral blood and their surface KIR receptors in women with endometriosis and healthy women using flow cytometry. Unfortunately, despite thorough analysis, we did not find any statistically significant differences between the studied groups. Further studies with larger sample sizes are needed to better capture biological variability and utilize a more comprehensive methodology for analysis.
