Genetic variation of selected pathogens in the house mouse hybrid zone

Abstract:

Parazitický způsob života je pravděpodobně nejrozšířenější životní strategií. Parazité se snaží využívat zdroje ze svého hostitele a ti se naopak snaží zdokonalovat mechanismy na svou obranu (např. imunitní systém), což vede k nikdy nekončícímu souboji zbraní. Na různých evolučních úrovních pak tento souboj vede k oboustranným adaptacím v důsledku selekčního tlaku parazitů na jejich hostitele (a naopak). V hybridních zónách (HZ) mezi dvěma diversifikovanými populacemi hostitelů, mohou být tyto adaptace snadno narušeny, což umožňuje v těchto dynamických systémech testovat nejrůznější hypotézy koevoluce mezi hostitelem a parazitem. Evoluční význam selekční dynamiky mezi hostitelem a parazitem v HZ je dosud málo prostudován, což je pravděpodobně důsledkem celkové složitosti tohoto systému. Hlavním náplní této disertace je přispět k pochopení koevoluce mezi hostitelem a parazitem, s využitím hybridní zóny jako přírodní laboratoře, k čemuž je využit detailní popis genetické variability a struktury vybraných parazitů odlišujících se některými znaky svých životních historií. Jako modelové skupiny byly zvoleny následující parazitické taxony: bakteriální rod Helicobacter a dva druhy ze skupiny Nematoda, Syphacia obvelata (Oxyurida) a Trichuris muris (Trichurida), izolované z myší domácích v oblasti intenzivně studované hybridní zóny mezi dvěma hostitelskými poddruhy ve střední Evropě. Hlavním cílem bylo porovnat genetickou variabilitu parazitů s genetickou strukturou jejich hostitelů (tj. rozdělení na dva poddruhy). Studované taxony parazitů byly vybrány s ohledem na jejich prevalenci, hostitelskou specificitu a další znaky životních historií, které mohou genetickou strukturu ovlivňovat. V první studii byla nalezena 100% prevalence rodu Helicobacter u volně žijících populací myší domácích, časté smíšené infekce více druhů Helicobacter a žádná zřetelná asociace druhového spektra s podruhy hostitele (všechny genetické linie byly běžně nalézány na obou stranách HZ). V následujících dvou studiích byla naopak nalezena velmi zřetelná genetická struktura silně specifického parazita Syphacia obvelata, která byla téměř identická s genetickou strukturou hostitele. Poprvé byli rovněž nalezeni hybridi mezi geneticky diversifikovanými liniemi S. obvelata. Tato zjištění byla v silném kontrastu s poslední studií, která analyzovala genetickou strukturou méně specializovaného druhu Trichuris muris. Tento druh parazita nevykazoval žádnou strukturu v závislosti na poddruhu hostitele. V disertační práci byly demonstrovány různé formy koevoluce mezi hostitelem a parazitem. Zároveň byly poskytnuty důkazy o tom, že variabilita ve vybraných znacích životních historií (např. specialisté vs. generalisté) má zcela zásadní vliv na genetickou strukturu populaci parazitu.

Abstract:

Parasitism represents probably the most widespread life-strategy. Parasites try to exploit the resources of their host and host attempt to maintain an efficient arsenal against intruding parasites (e.g. immune system), which leads to never ending host parasite arms-race. At macro- and microevolutionary level it forms the basis of numerous spatially variable local adaptations due to selection pressure imposed by parasites upon their hosts and vice versa. In hybrid zones (HZ) between two divergent host populations, where hosts with novel recombinant genetic architecture emerge, these adaptations may break down. This fuels the race of evolution between host and parasite and provides a ground to test several evolutionary hypotheses by studying this dynamic system. The evolutionary significance of this ‘parasite-host dynamics’ is poorly understood, partly because it is so complex. The main objective of my PhD thesis was to enhance understanding of host parasite coevolution using hybrid zone as a natural laboratory broadly, and to infer the spatial population structuring and genetic variability in selected parasites species keeping parasite life history traits as variables specifically. This PhD work aimed to study genetics of following selected parasites: one bacterial genus Helicobacter and two nematode species, Syphacia obvelata (Oxyurida) and Trichuris muris (Trichurida), collected from mice from the HZ of two subspecies of the house mouse in Central Europe. The main aim was to compare genetic variability of parasites with the genetic background of their hosts. Studied parasitic species were chosen according to their prevalence, specificity and other life history traits towards host as these can theoretically influence their genetic structure. Firstly, I described that the prevalence of Helicobacter in the house mouse is 100%, mixed infections of several species are very common, and there is no clear relationship with host genetic background as all genetic lineages are common on both sides of HZ. In the next studies, I showed clear genetic structure of a very specific nematode parasite of the house mouse, Syphacia obvelata, in strong correlation with host genetic background. I also evidenced for the first time the presence of hybrids of Syphacia in the mouse hybrids. On the contrary, in more generalist nematode Trichuris muris, no signs of genetic structure were observed with respect to the host subspecies. In conclusion, we demonstrated ultimate form of host-parasite coevolution and furthermore actualized the idea that variability in parasite life history traits such as specialism vs generalism influences their genetic variability and spatial population structure.
