Toxicity mechanisms in early developmental stages of aquatic animals

Abstract:

V souvislosti s masovým rozvojem a negativním vlivem vodního květu na živé organismy roste potřeba studia účinků a mechanismů toxicity sinic. Jedním z málo zkoumaných vlivů je možná embryotoxicita a teratogenita cyanobakterií a jejich toxinů. Pro odhad rizika embryotoxicity a teratogenity chemických látek a environmentálních směsí se v ekotoxikologii používá standardizovaný test využívající embrya drápatky vodní (FETAX). Tato práce je zaměřena na vliv jednotlivých frakcí vybraných laboratorních monokultur sinic (Aphanizomenon klebahnii, Aphanizomenon gracile, Microcystis aeruginosa) a řas (Scenedesmus quadricauda, Chlorella kessleri) na raný vývoj embryí Xenopus laevis a úlohu deficitu kyslíku během testů FETAX. U výše uvedených monokultur byl testován vzorek komplexní biomasy, která byla centrifugací rozdělena na dvě další frakce - buněčný debris (u sinic obsahující lipopolysacharidy) a supernatant. V průběhu testu byl měřen obsah rozpuštěného kyslíku jako důležitý parametr testování. Ze získaných výsledků je zřejmé, že toxicita vyvolaná expozicí není způsobena pouze přítomností cyanotoxinů ve vzorku. Škodlivé účinky měly i vzorky připravené z biomas zelených řas. Obsah rozpuštěného kyslíku v testované směsi se ukázal jako důležitý faktor ovlivňující přežití a fitness embryí.

Abstract:

Mass development of water bloom and its negative effects on organisms in aquatic environment lead to growing demand for research of toxic mechanisms and effects induced by cyanobacteria. Embryotoxicity and teratogenity of cyanobacteria and its toxins have not been widely studied yet. In the present study, we have investigated toxic effects of several laboratory cultures of cyanobacteria (Aphanizomenon klebahnii, Aphanizomenon gracile and Microcystis aeruginosa) and green algae (Scenedesmus quadricauda and Chlorella kessleri) on the development of Xenopus laevis using standardized FETAX. Tested samples included complete algal/cyanobacterial biomass; cell debris (containing e.g. cell bound lipopolysacharides) and supernatant after centrifugation of complex biomass. We have also monitored oxygen concentrations as they were previously shown to be an important parameter in the ecotoxicity testing of cyanobacterial blooms. The obtained research data indicate that the conditioning-induced toxicity is not caused only by the presence of cyanobacteria in the sample. Also the samples from algal biomass had toxic effects. In our findings, the amount of dissolved oxygen in the tested sample proved to be an important agent affecting embryo survivability and fitness.
