Natural antibodies against a-Gal epitope: development of enzyme immunoassay and applications in clinical immunology

Abstract:

Byla vyvinuta původní enzymová imunoanalytická metoda (ELISA), která umožňuje kvantitativní stanovení přirozených antigalaktosylových protilátek (anti-Gal) v isotypech IgG, M, a A. Tyto protilátky jsou přítomny v séru všech zdravých osob, jejich koncentrace je interindividuálně variabilní. Statisticky významně vyšší koncentraci anti-Gal IgM mají ženy, anti-Gal IgA muži. U osob s krevní skupinou B je hladina anti-Gal ve všech isotypech signifikantně nižší než u ostatních krevních skupin. Tvorba anti-Gal v prvních dvou letech života koreluje s tvorbou celkových IgM, IgG a IgA, v pupečníkové krvi jsou prokazatelné pouze anti-Gal IgG. V séru pacientů s primárními hypogamaglobulinemiemi ve srovnání se zdravými osobami jsou anti-Gal protilátky přítomny ve významně nižších koncentracích: u pacientů s běžnou variabilní imunodeficiencí ve všech třídách imunoglobulinů, u pacientů se selektivním deficitem IgA pouze v isotypu IgA a u pacientů se selektivním deficitem IgM nejvýrazněji v IgM, ale statisticky významně i v IgG a v IgA. Pacientky s mamárním karcinomem a pacienti, muži, s kolorektálním karcinomem mají ve srovnání se zdravými signifikantně nižší koncentrace anti-Gal IgM. Implantace chlopní a aortokoronární bypass ovlivňují hladiny protilátek anti-Gal: první den po operaci dochází k jejich snížení, za 6–11 dnů byl zaznamenán vzestup, který byl nejvýraznější u příjemců chlopní biologických v IgA.

Abstract:

The original enzyme immunoassay (ELISA) for quantitative determination of naturally occurring anti-galactosyl (anti-Gal) antibodies in the IgG, IgA and IgM isotypes was developed. These antibodies are present in serum of healthy population with inter-individually variable concentration. Women have statistically significant higher levels of IgM anti-Gal antibodies, on the other hand men have statistically significant higher levels of IgA anti-Gal antibodies. With regard to the blood groups, people with group B have statistically significant lower concentrations of anti-Gal antibodies in all three measured isotypes compared to other main blood groups. Production of IgG, IgA and IgM anti-Gal antibodies correlates with secretion of IgG, IgA and IgM immunoglobulin isotypes; only IgG anti-Gal antibodies are present in cord blood. The patients with primary hypogammaglobulinemia have statistically significant lower levels of anti-Gal antibodies in serum; patients with common variable immunodeficiency have decreased concentration of IgG, IgA and IgM isotypes and patients with selective IgA deficiency decreased concentration of IgA isotype of anti-Gal antibodies; patients with selective IgM deficiencies have markedly decreased level of IgM class of anti-Gal antibodies accompanied by statistically significant decreased level of IgG and IgA class of anti-Gal antibodies. Women suffered from breast cancer and men suffered from colorectal carcinoma have statistically significant lower levels of IgM anti-Gal antibodies compared to healthy individuals. Aortic valve implantation and aortocoronary bypass influence the levels of anti-Gal antibodies: decrease of anti-Gal antibodies on the first postoperative day was accompanied by increase of anti-Gal antibodies during 6 to 11 days after transplantation which was the most significant in IgA anti-Gal antibodies in the recipients of biological aortic valves.
