Detection of N-glycan structures isolated from serum and tissues of patients suffered with breast cancer

Abstract:

Glykomika je rozsáhlá vědní disciplína zkoumající strukturu a vlastnosti glykanů, monosacharidů a oligosacharidů kovalentně navázaných na jiné biomolekuly, především proteiny. Glykany jsou zapojeny do mnoha důležitých procesů v buňce jako je mezibuněčná komunikace, proliferace nebo lokalizace a skládání proteinů. Bylo prokázáno, že u nádorových buněk dochází ke změnám v N-glykanovém profilu, především ke zvýšení hladin polysialylovaných, větvených, komplexních glykanů. Tyto glykany hrají velmi důležitou roli při růstu a vývoji nádoru, angiogenezi a metastazování. Diplomová práce je součástí projektu GAČR 16-04496S; 2016–2018 s názvem: „Glykomické profilování se vztahem k biomolekulám rozlišujícím nádory prsu“ a je zaměřena na profilování N-glykanových struktur izolovaných ze vzorků sér a tkání pacientek s diagnostikovaným karcinomem prsu. V první části experimentální práce byly optimalizovány procesy izolace a purifikace N-glykanových struktur, druhá část práce byla zaměřena na porovnání N-glykanových profilů sér a tkání pacientek s nádory prsu odlišných molekulárních subtypů (HER2 pozitivní a luminal A). V tkáních i sérech byly nalezeny N-glykany se statisticky významně rozdílnou hladinou (p<0,05) mezi vybranými subtypy. Projekt bude dále zaměřen na profilování N-glykanů izolovaných ze sér a tkání ostatních subtypů karcinomu prsu, výsledky budou evaluovány na vybraných buněčných liniích odvozených od daných subtypů.

Abstract:

Glycomics is a an extensive field of science focused on the structure and function of glycans, monosacharides and oligosacharides covalently bound to other biomolecules, especially proteins. Glycans are involved in many important processes in cell like cell-to-cell communication, proliferation or localization and folding of the proteins. It has been demonstrated that cancer cells have changed N-glycan profiles, especially increased levels of polysialylated, branched, complex glycans were described. These glycans play a very important role in the progression of the tumour, angiogenesis and metastasis. The thesis is a part of the project GAČR 16-04496S; 2016–2018 called: „Glycobiological investigations pertaining to biomolecular differentiation of mammary carcinomas“ and is focused on the profiling of N-glycan structures isolated from the serum and tissue samples from patients with breast carcinomas. In the first part of the experimental work, the processes of isolation and purification of N-glycan structures were optimized, the second part was focused on the comparison of the N-glycan profiles of serum and tissues from patients with breast tumours with different molecular subtypes (HER2 positive and luminal A). In both tissue and serum, N-glycans with significantly different levels (p<0,05) were found between selected subtypes. In the future, the project will be focused on the profiling of N-glycans isolated from the serum and tissues of other subtypes of breast carcinomas, the results will be evaluated on the chosen cell lines derived from given subtypes.
