Phylogeography, population genetic structure and potential hybridization of Ethiopian endemic rodents of the genus Stenocephalemys

Abstract:

The Stenocephalemys endemic rodent genus includes four recently known species and one potentionally new species in the area of Ethiopian High Plateau. There is a pattern of altitudial gradient distribution among these species from the typical forrest habitat prefference (S. albipes) of lower altitudes, through heathland biotope species (S. griseicauda) of the middle altitudes and high-altitudes specialists (S. albocaudata, „species A“) in afro-alpine biotopes at the top. S. ruppi, that is morphologically intermediate, was excluded of the analysis for the probability of its extinction. The aim of this thesis was to make a review of phylogenetic relationships between these species, to analyze the population-genetic structure in the context of phylogeography and to test the potentional hybridization in the species contact zone in Bale Mountains. Mitochondrial gene sequences of cytochrome b (139 specimens) and 7 nuclear microsatelites loci (324 specimens, 28 populations) was used for the analysis. Nuclear loci clearly distinguished all species. Mitochondrial DNA analysis also supported the species and clarified phylogenetic relationships. Furthermore, the analysis outlined a bigger interpopulational structure of this taxa. Mainly S. griseicauda was segmented into two genetically different clades splitted by the Great Rift Valley as it was seen in the other species. The separation of S. albipes into two clades is noticeable at the base of mt DNA where the south clade B includes two series of unique haplotypes from the localities of the Inegawa Forrest and Chercher Mountains and also one specimen with a unique haplotype which localization is on one of the Tana Lake islands near the North clade A. This segmentation was not very well supported by microsatelite loci. The question of hybridization in the Bale Mountains has still remained open.

Abstract:

Endemický rod hlodavců Stenocaphalemys zahrnuje 4 doposud popsané druhy a 1 potenciálne nový druh v oblastech Etiópske náhorni plošiny. V distribuci jednotlivých druhů můžeme vidět trend na základe altitudiálniho gradientu od typicky lesního druhu (S. albipes) nižších nadmorských výšek, cez druh obývajíci vresoviště středných poloh vysočiny (S. griseicauda), až po vysokohorských specialistů na afro-alpínsku tundru (S. albocaudata, „species A“). S. ruppi., který vytváří morfologický medzistupeň lesního a horských druhů, byl z této práce vynechán z důvodu pravdepodobné extinkcie. Cílem této práce bylo revidovať fylogenetické vztahy druhů, analyzovat populačně genetickú strukturu ve fylogeografickom kontextě a testovať hypotézu možné hybridizace v oblasti kontaktných zon 3 druhů v Bale Mountains. Použité boli sekvence mitochondriálniho genu pro cytochróm b ze 139 jedincov a 7 mikrosatelitových lokusů pro 324 jedincov z 28 populácí. Na základě jadrových znaků byli jednoznačne odlíšené druhy. Tí byli taktéž podpořené analýzou mtDNA, která objasnila rovněž ich fylogenetické vztahy. Analýzy načrtli taktéž vyšší strukturovanosť týchto taxónů. Hlavně S. griseicauda byla rozčlenena na 2 geneticky odlišné klády oddelené Velkou překopovou propadlinou, jak možná pozorovat taky u jiných druhů. Značné je členení S. albipes na základě mtDNA na dvě skupiny, kde jižný klád B zahnuje 2 série unikátnich haplotypů z lokalít Inegawa Forrest a Chercher Mountains a taktéž jedince s unikátnim haplotypem, kterého lokalizace je na jedném z ostrovů jazera Tana v blízkosti skupin severního kládu A. Táto segmentáce nebyla jednoznačně podpořena mikrosatelitovými lokusmi. Otázka hybridizace v Bale Mountains zůstává nadále otevřená.
