Oxidative stress in denitrifying bacteria

Abstract:

The nitrogen cycle in nature is in no small part influenced by denitrification bacteria. The model organism for studying this process is Paracoccus denitrificans. Due to the fact that it is an obligatory respirating bacteria, its metabolism is under strain from oxidative stress. In a previous research by Pernkířová et al. 2015 this bacteria was put under increased oxidative stress and a following proteomic analysis showed elevated levels of several proteins. On the basis of these results three mutant strains were created, RegA-, RegB- a 5119-. In this study the strains were exposed to different concentrations of an agent inducing oxidative stress and their growth was monitored turbidimetrically. In the case of mutats RegB- and 5119- a significant decrease or even halt in growth compared to the wild strain was shown, the growth of RegA- couldn´t be measured properly due to technical difficulties. A study of growth on different carbon substrates was also conducted. It was discovered that the strains RegB- and 5119- could not effectively utilize methanol and butyrate substrates as a carbon source.

Abstract:

Cyklus dusíku v přírodě je z nemalý míry ovlivňován denitrifikačními bakteriemi. Modelovým organismem studia tohoto procesu je Paracoccus denitrificans. Vzhledem na to, že se jedná o obligátně respirující bakterii, je její metabolismus zatížen oxidačním stresem. Ve předešlém výzkumu Pernkířová et al. 2015 byla tato bakterie vystavena zvýšenému oxidačnímu stresu a následní proteomická analýza ukázala zvýšenou hladinu několika proteinů. Na základe těchto výsledků byli vytvořeny tři mutantní kmeny, RegA-, RegB- a 5119-. Ty byli v tomhle studiu vystaveny různým koncentracím činidla indukujícího oxidační stres a jejich růst sledován turbidimetricky. V případě RegB- a 5119- došlo ku výraznému zpomalení až zastavení růstu oproti divokému kmenu, růst RegA- se z technických příčin nepodařilo věrohodně zaznamenat. Rovněž byl realizován růst na různých uhlíkatých substrátech. Při kmenech RegB- a 5119- se zjistilo, že nedokážou efektivně využít methanol a butyrát jako zdroj uhlíka.
