Abstract:

V práci zaměřené na kultivaci želvušek byly zkoumány vlivy různých kultivačních médií na životaschopnost zásobních buněk želvušek a samotných želvušek. Želvušky byly izolovány z mechorostů z lokalit Ústí nad Orlicí, Hamr na Jezeře a Sloup v Čechách pomocí extrakční techniky s využitím sítka. Hlavním cílem práce bylo pozorovat, jak jednotlivá kultivační média ovlivňují životaschopnost zásobních buněk želvušek. K tomuto účelu bylo testováno devět médií. Zároveň byl sledován vliv těchto médií na životaschopnost samotných želvušek. Výsledky ukázaly, že v všech devíti testovaných médiích byly nalezeny živé želvuščí buňky. Z toho lze usoudit, že všechna zkoumaná média poskytovala potřebné živiny a podmínky pro přežití želvušek. Dále bylo zjištěno, že osm z těchto devíti médií podporovalo pohyb želvušek. To naznačuje, že tato média měla pozitivní vliv na vitalitu a aktivitu želvušek.

Abstract:

In the work focused on the culture of tardigrades, the effects of different culture media on the viability of stock cells and the tardigrades themselves were investigated. The tardigradess were isolated from bryophytes from the localities of Ústí nad Orlicí, Hamr na Jezeře and Sloup in Bohemia using a sieve extraction technique. The main aim of the work was to observe how different culture media affect the viability of the tardigrades storage cells. Nine media were tested for this purpose. At the same time, the effect of these media on the viability of the tardigrades themselves was observed. The results showed that viable tardigrades cells were found in all nine media tested. It can be concluded that all the media tested provided the necessary nutrients and conditions for the survival of the tardigrades. Furthermore, eight of these nine media were found to support tardigrades movement. This suggests that these media had a positive effect on the vitality and activity of the tardigrades.
