Determination of selected phytocannabinoids with liquid chromatography

Abstract:

The increasing interest in cannabinoids makes it a necessity to state their content in the plant material for standard practice. The HPLC method features low LOD and LOQ without the need of a complex sample preparation. The HPLC system has been used with a reversed configuration and testing of various mobile phases (acetonitrile:buffer, methanol:buffer), the composition of the buffer (ammonium formate, ammonium acetate), pH of the buffer (3.8 – 5.2) and its concentration (0.1% and 0.2%), the gradient, columns (Thermo Scientific Accucore C18 100mm*3mm*2.6µm, Sigma-Aldrich Ascentis Express C18 100mm*3mm*2.7µm) and the columns temperature (25-55 °C). Additionally we tested the extraction from raw plant material mixed with three extraction solvents (methanol:chloroform, isopropanol:chloroform, hexane:butanol) used for extraction. The required sensitivity with properly separated peaks (LOQ- CBDA 87.06 ng/ml, CBD 174.3 ng/ml, THCA 73.25 ng/ml a THC 54.5 ng/ml) was achieved with the Accucore C18 column and the mobile phase of ammonium formate/methanol (0.1%, pH 4.2) with the initial gradient of 72% methanol at the temperature of 50 °C. Mixture of methanol:chloroform 9:1 was chosen for extraction with 80.1% recovery.

Abstract:

S narůstajícím zájmem o kanabinoidy je nutností stanovit jejich obsah v rostlinném materiálu v běžné praxi. HPLC se vyznačuje nízkými limity kvantifikace bez náročné přípravy vzorku. Při vývoji metody byl použit systém HPLC s reverzním uspořádáním a byly testovány různé mobilní fáze (acetonitril:pufr, metanol:pufr), složení pufru (mravenčan amonný, octan amonný), pH pufru (3,8 – 5,2) a jeho koncentrace (0,1% a 0,2%), gradient, kolony (Thermo Scientific Accucore C18 100mm*3mm*2,6µm, Sigma-Aldrich Ascentis Express C18 100mm*3mm*2,7µm) a teplota kolony (25-55°C). Taktéž byla testována extrakce z rostlinného materiálu se třemi extrakčními činidly (metanol:chloroform, izopropanol:chloroform a hexan:butanol). Požadovaná citlivost s dobře separovanými píky a limity kvantifikace (LOQ) pro CBDA 87,06 ng/ml, CBD 174,3 ng/ml, THCA 73,25 ng/ml, THC 54,5 ng/ml) byla dosažena s kolonou Accucore C18 a mobilní fází mravenčan amonný/metanol (0,1%, pH 4,2) s počátečním gradientem 72 % metanolu při teplotě 50 °C. Pro extrakci byla zvolena směs organických rozpouštěděl methanol:chloroform 9:1 s výtěžnosťí 80,1 %.
