Cell cycle checkpoints and DNA repair in relation to anti-cancer therapy

Abstract:

Odpověď buňky na poškození DNA (tzv. DNA damage response; DDR) zahrnuje koordinovanou reakci proteinů zastavujících buněčný cyklus v klíčových kontrolních bodech, zejména G1/S a G2/M, a dále reparaci poškozené DNA. Pokud tato reparace není možná, spouští buňka programovanou buněčnou smrt – apoptózu. Na indukci apoptózy je také založena (téměř) veškerá protinádorová terapie zahrnující cytostatika a ionizující záření. Tato modelová situace je komplikována mutacemi a genomickými aberacemi v důležitých genech účastnících se DDR. Bakalářská práce shrnuje poznatky o kontrolních bodech buněčného cyklu a možnostech zablokování oprav DNA, jakožto prostředku protinádorové terapie. Práce je zaměřena zejména na koncept syntetické letality.

Abstract:

Cellular response to genotoxic stress (DNA damage response, DDR) involves a coordinated reaction of proteins, which regulate cell cycle at the key checkpoints, namely G1/S and G2/M, and also ensure a repair of damaged DNA. If the damage is too extensive, a cell can elicit programmed cell death – apoptosis. Induction of apoptosis through appropriate DNA damage represents a cornerstone of almost all anti-cancer therapy, which includes mostly chemotherapy and irradiation. This model situation is complicated by mutations and genomic aberrations in genes important for the DDR. Bachelor thesis covers the key findings concerning cell cycle checkpoints and outlines some possibilities how to inhibit DNA repair as a way of anti-cancer therapy. The thesis is focused particularly on the concept of so-called synthetic lethality.
