Selection of the Optimal Method For Embryo Cryopreservation

Abstract:

Bakalářská práce se zaměřuje na srovnání kryokonzervačních technik, které se využívají v centrech asistované reprodukce. Nadbytečná embrya vzniklá mimotělním oplodněním je možno kryokonzervovat pro pozdější využití buď metodou vitrifikace nebo pomalým mražením. Kryokonzervaci lze provést v časném vývoji embrya, tedy v prvních šesti dnech po oplození. Práce srovnává metody mražení u vybraných stádií embrya (zygot ve stadiu prvojader, dělících se embryí a blastocyst) v následujících parametrech: v počtu přežitých embryí po rozmražení, v míře úspěšnosti klinického těhotenství a v porodnosti. Nejdůležitějším parametrem pro hodnocení úspěšnosti kryokonzervačních technik je míra porodnosti, která zaznamenala ve srovnávaných studiích lepší výsledky při metodě pomalého mražení ve všech třech stádiích. Zároveň je třeba brát v úvahu, že studie byly nejednotné v řadě vstupních determinantů (věk, typ kryoprotektiv, aj.), které mohly konečné výsledky ovlivnit. Dokud se výzkumy zabývající se danou problematikou nesjednotí v těchto faktorech, nelze určit, která z metod je pro kryokonzervaci vhodnější.

Abstract:

This thesis focuses on the comparison of cryopreservation techniques which are used in assisted reproduction centers. Superplus embryos resulting from in vitro fertilization can be preserved by freezing for later use, either by vitrification or slow freezing. Cryopreservation can be performed in early development of the embryo, which takes place in the first six days after fertilization. The thesis compares different methods of freezing the embryo in selected stages (pronuclear zygotes, cleavage-stage embryos and blastocysts) in the following parameters: the number of embryos survived after thawing, the success rate of clinical pregnancy, and birth. The most important parameter for evaluating the success of cryopreservation techniques is birth rate. As it was shown in all compared studies, the slow freezing method led to the best results in this parameter in all three stages. However, it should be borne in mind that the studies were inconsistent in a number of entry determinants (age, type of cryoprotectants, etc.) that could affect the final results. Therefore, we can not unreservedly determine the most viable method of cryopreservation until further unification of the respective determinants is made.
