Analysis of functional importance of C1 inhibitor promoter mutations

Abstract:

Hereditární angioedém je onemocnění způsobené nedostatkem funkčního C1 inhibitoru. U pacientů s tímto onemocněním byly do dnešního dne popsány tři promotorové mutace, jejich vliv na funkci promotoru zatím nebyl prokázán. V této diplomové práci je zkoumán vliv promotorových mutací genu pro C1 inhibitor na regulaci transkripce genu. V rámci této práce byla v genetické laboratoři CKTCH zavedena metodika analýzy funkce promotoru pomocí reportérských genů, luciferáz. Tato metoda umožnila posoudit vliv promotorových mutací porovnáním aktivity mutantních promotorů s aktivitou promotoru divokého typu. Získané výsledky ukázaly, že dvě ze tří zkoumaných mutací, -40 C>G a -45 C>A, aktivitu promotoru pravděpodobně vůbec neovlivňují. Třetí mutace, tranzice -103 C>T, zvyšovala expresi reportérského genu ze zkoumaného úseku promotoru téměř o 60%. Příčina snížené hladiny C1 inhibitoru u pacientů s touto mutací není zřejmá. Jednou z možností je, že existuje negativní korelace mezi množstvím mRNA v buňkách a množstvím C1 inhibitoru v krevním séru. Taková negativní korelace již byla popsána, regulační mechanizmus, který se v tomto procesu uplatňuje, však zatím není znám.

Abstract:

Hereditary angioedema is caused by the autosomal-dominant deficiency of C1 inhibitor gene. Till now, three promoter mutations of the gene have been detected. However, the impact of these mutations on promoter function has not been uncovered yet. In this study, I present results of investigation of functional importance of the three promoter mutations for the transcription of C1 inhibitor gene. During this investigation, luciferase reporter assay has been established in genetic laboratory CKTCH. This methodology enabled me to compare the promoter activity of mutated and non-mutated promoters. Two of the three promoter mutations, -40 C>G a -45 C>A, do most likely not influence the function of C1 inhibitor promoter. The mutation -103 C>T, contrary to the previous ones, increased expression of reporter gene by 60 % in comparison to non-mutated promoter. However, plasma levels of C1 inhibitor in patients with this mutation are significantly decreased. The reason for this discrepancy is not obvious. Maybe there is a negative correlation between amount of mRNA in the cells and plasma levels of C1 inhibitor. Such a correlation has been described in healthy controls. Nevertheless, its mechanism is yet unclear.
