The role of CD36 receptor in the perception of taste of fatty acids

Abstract:

Receptor CD36 je široce rozšířeným membránovým proteinem, který je schopen vázat mnoho ligandů včetně proteinů či patogenů. Vyskytuje se v různých typech buněk a jeho přítomnost byla prokázána také v chuťových buňkách hrazených papil na jazyku. Zde je schopen vázat mastné kyseliny s dlouhými řetězci a přepravovat je do buňky, kde následuje kaskáda dějů, které vyústí k signalizaci nervového vzruchu z chuťových pohárků do centrální nervové soustavy. Percepce chuti mastných kyselin může být ovlivněna mnoha faktory. V současné době se největší pozornost věnuje genu pro receptor CD36 a jeho polymorfismům, které mohou mít vliv na funkci tohoto receptoru. Cílem této bakalářské práce byl nábor 53 osob z české populace, provedení chuťového testu na rozpoznávání jednotlivých koncentrací kyseliny linolové, testu preferencí potravin a změření některých antropometrických údajů. Dalším z cílů práce byla analýza odebrané DNA na vybraný polymorfismus genu pro CD36. Po statistickém vyhodnocení nebyly nalezeny žádné významné propojení mezi polymorfismem rs1761667, antropometrickými parametry a prahy detekcí. Součástí práce bylo také zavedení metodiky pro stanovení exprese genu pro CD36 na jazyku myši. Zavedená metodika se bude v budoucnu využívat při stanovení exprese genu pro CD36 na jazyku myši v závislosti na rozdílné potravě.

Abstract:

CD36 receptor is a widely spread membrane protein which it is capable of binding many ligands including proteins or pathogens. It is present in many types of cells and its presence has also been demonstrated in the taste buds of circumvallate papillae on tongue. There, it is able to bind long chain fatty acids and transport them into the cells, followed by a cascade of events that leads to nerve signaling from the taste buds to the central nervous system. The perception of taste of fatty acids can be influenced by various factors. Nowadays, the most studied aspect is the CD36 gene and its polymorphisms, which may affect the function of this receptor. The aim of this bachelor thesis was to test 53 persons from the Czech population, whether they can identify individual concentrations of linoleic acid, their food preferences and to measure some anthropometric data. Furthermore we analyzed the collected DNA for the selected polymorphism of the CD36 gene. Anther contribution of this thesis is the introduction of statistical evaluation, there was no significant link between the rs1761667 polymorphism, anthropometric parameters and threshold detection. Part of the thesis was also the introduction of methodology for determination of CD36 gene expression on mouse tongue. The established methodology will be used in the future to determine the expression of CD36 gene on the tongue of the mouse depending on different foods.
