Microbial occurrence in technology of paper production

Abstract:

S průmyslovými vodními systémy je spojena řada mikrobiologických problémů. Tyto problémy jsou spojeny především s růstem mikroflory, tj. směsí různých řas, hub a bakterií. Jedním z průmyslových odvětví s velkou spotřebou technologické vody je papírenský průmysl. V těchto vodách je vhodné prostředí pro množení mikroorganismů a právě zde by nekontrolovatelný růst mohl způsobovat velké škody např. zvýšením energetických nákladů nebo poškozením strojního zařízení. To by mělo dopad jak na životní prostředí, tak i na kvalitu finálního produktu. V rámci této diplomové práce byla zjišťována závislost růstu hygienicky významných skupin mikroorganismů na ročním období kultivací na selektivních pevných půdách. Z pokusů vyplynulo, že růst mikroorganismů ročními obdobími výrazně ovlivněn nebyl, což je způsobeno také tím, že v rámci provozu je udržována stála teplota technologické vody. Další částí diplomové práce byla identifikace vybraných skupin bakterií (čeleď Enterobacteriaceae a skupina nefermentujících tyček). Neznámé kmeny byly identifikovány pomocí komerčních i konvenčních sad testů. Pro zpřesnění a porovnání výsledků bylo využito i chemotaxonomické metody MALDI MS. Mezi identifikovanými kmeny nebyl zjištěn žádný patogen ve zvýšené frekvenci.

Abstract:

The industrial water system is connected to a number of microbiological problems. These problems are mainly related to the growth of bacteria, namely mixtures of different algae, fungi and bacteria. One of the industries with high consumption of water is a paper technology based industry. In these waters there is a suitable environment for microbiological growth and this rampant growth could cause major damage such as increased energy costs or damage on machinery. This would affect both the environment and the quality of the final product. One part of this thesis is devoted to establishing dependence of growth of hygienically significant groups of microorganisms on the season by cultivation on selective solid soils. The experiments showed that the growth of microorganisms was not significantly affected by season. This may be due to the fact that constant temperature of process water is maintained. Another part of the thesis was to identify selected groups of bacteria (family Enterobacteriaceae and group of nonfermenting rods). Unknown strains were identified using commercial kits and conventional tests. For a more precise comparison of the results a chemotaxonomic method MALDI MS was used. Among the identified strains, no pathogen was detected in increased frequency.
