Imune response of chickens to Salmonella infections

Abstract:

Salmonella enterica je významným střevním patogenem člověka i hospodářských zvířat. U člověka ale i drůbeže je prvotní odpovědí imunitního systému střevního traktu na infekci S. enterica exprese široké škály chemokinů a prozánětlivých cytokinů, které jsou secernovány jak epiteliálními buňkami tak i buňkami imunitního systému, především makrofágy. Funkce většiny prozánětlivých cytokinů však byla charakterizována u jiných modelových zvířat než u drůbeže a u kura domácího byla existence těchto cytokinů často jen předpovězena na základě sekvenování genomu a porovnání identifikovaných genů se sekvencemi v genových buňkách. Protože bazální exprese prozánětlivých cytokinů v různých orgánech drůbeže není v podstatě známa, ve své bakalářské práci jsem se zabývala studiem exprese vybrané skupiny 12 cytokinů (IL-1β, IL-12p35, IL-12β, IL-15, IL-17, IL-18, IL-22, IL-8, IFNβ, IFNγ, TNF-α a iNOS) u 33 týdnů starých slepic kvantifikací mRNA pomocí Real-time-PCR. Expresi jsem stanovila ve 4 částech gastrointestinálního traktu (ileum, ileocaecální chlopeň, tlusté a slepé střevo). Exprese všech sledovaných cytokinů ve všech částech střevního traktu byla velmi podobná. Nejvyšší exprese byla pozorována u TNF-α a IL-15 jejichž exprese byla 10x až 100x než u zbylých cytokinům. Mezi nejméně exprimované cytokiny ve střevní traktu patřil IL-12p35, jehož exprese ve slepém střevu byla pod detekčním limitem Real-time-PCR. Dosažené výsledky mohou být využity jako výchozí hladiny pro stanovení exprese po infekci patogenními mikroorganismy včetně S. enterica.

Abstract:

Salmonella enterica is an important enteric pathogen in humans and livestock. In humans, as well as poultry is the primary immune response to intestinal infection, S. enterica expression of a wide range of chemokines and proinflammatory cytokines that are secreted as epithelial cells and cells of the immune system, particularly macrophages. The function of most pro-inflammatory cytokines, however, was characterized in other model animals than poultry and domestic fowl, the existence of these cytokines often predicted on the basis of genome sequencing and comparison of identified genes with sequences in gene cells. Because the basal expression of proinflammatory cytokines in various organs of poultry is essentially unknown, in my thesis I dealt with the study of expression of selected groups of 12 cytokines (IL-1β, IL-12p35, IL-12β, IL-15, IL-17, IL- 18, IL-22, IL-8, IFNβ, IFNγ, TNF-α and iNOS) at 33 weeks old hens quantification of mRNA using Real-time-PCR. Expression I set out in 4 parts of the gastrointestinal tract (ileum, ileocaecal flap, colon and the appendix). Expression of all investigated cytokines in all parts of the intestinal tract were very similar. The highest expression was observed in TNF-α and IL-15 whose expression was 10 times to 100 times before the remaining cytokines. Among the least expressed cytokines in the intestinal tract was IL-12p35, the expression of the appendix was below the detection limit of Real-Time-PCR. Achievements can be used as the default setting for the expression levels after infection with pathogenic microorganisms, including S. enterica.
