Abstract:

Úpravy povrchů materiálů vhodných pro kontakt s tělními tkáněmi jsou stále více předmětem zájmu pro jejich potenciální využití v tkáňovém inženýrství. Jednou z možných modifikací je vystavení materiálu účinkům plazmatu elektrického výboje v parách cyklopropylaminu. V tomto typu plazmatu dochází k plazmové polymerizaci, která vytvoří tenkou povrchovou vrstvu obsahující aminové skupiny. Předpokládá se, že tento povrch je vhodný pro adhezi a proliferaci buněk. Cílem diplomové práce bylo tento předpoklad ověřit a zjistit, jaké depoziční podmínky cyklopropylaminových vrstev jsou nejvhodnější. Buňky myších myblastů C2C12 rostoucí na plazmatem upravených površích proliferovaly srovnatelně s kontrolními neupravenými povrchy, ale lišily se abnormálně vysokou úrovní adheze. Za účelem odhalit příčinu těchto jevů byla na dvou různých variantách povrchu sledována úroveň exprese proteinů zapojených do těchto procesů. Exprese proteinů podílejících se na formování adherentních spojů, fokálních adhezí a stresových vláken se mezi kontrolními buňkami a buňkami na experimentálních površích nelišila. Buňky též nepodléhají řízené buněčné smrti a jejich buněčný cyklus je srovnatelný s buněčným cyklem buněk kontrolních. Rozdílná exprese byla detekována pouze v případě proteinů fibronectin, tubulin III β a lamin A/C, což naznačuje, že by mohlo docházet k iniciaci procesu diferenciace. Tento závěr je však nutné potvrdit dalšími experimenty.

Abstract:

Surface modification of the materials, which are suitable for contact with tissues, are in past few years object of the interest for their potential use in tissue engineering. One of the possible surface modifications is the exposition to the plasma electric discharge in cyclopropylamine vapors. This specific type of exposition leads to process called plasma polymerization, which results in thin layers containing amine groups. It is believed that this type of surface modification is suitable for the adhesion and proliferation of cells. The aim of this thesis was to verify above mentioned hypothesis, and moreover, to identify which conditions for deposition of cyclopropylamine layers are the best. For all the experiments we used the murine myoblast cell culture C2C12. After exposure of the cells to modified surfaces, they proliferated at the same level as cells on unmodified culture petri dishes. When we checked the strength of the adhesion, we found out that adhesion was greater on the modified ones. In order to discover the cause of this phenomenon, we checked levels of the expression of specific proteins, which could be important in the process of adhesion. Results showed that there is no difference in the expression of the proteins involved in the formation of focal adhesions, stress fibers and adherent junctions between cells on control and modified surfaces. Cells did not undergo apoptosis and there were not differences in the cell cycle as well. The only differences we found were in the expression of the proteins fibronectin, lamin A/C and tubulin β III, implying changes associated with differentiation. This conclusion, however, must be confirmed by further experiments.
