First steps towards the introduction of reverse genetics methods in the eustigmatophyte alga Vischeria

Abstract:

Práce se zabývá eustigmatofyty jakožto modelovými organismy molekulární biologie, a to se zaměřením na kmen Vischeria sp. C074, jehož základní charakteristika a biologie (např, růst či senzitivita na antibiotika) je klíčová pro následné využití jakožto modelového eustigmatofyta. Tato bakalářská práce se skládá ze dvou části, teoretické a praktické. Teoretická část je zaměřená na charakteristické rysy řasové skupiny eustigmatofyta její klasifikaci zahrnující historii a současnost této skupiny, praktickému využití se zaměřením na významné zástupce Nannochloropsis oceanica a Visheria. Dále tato práce shrnuje metody molekulární biologie využívané pro genetickou modifikaci organismů s konkrétními příklady využití právě na eustigmatofytech. Praktická část se zaměřuje na získaní základních informací, které je nutné stanovit před modifikací organismu. Zahrnuje stanovení růstové křivky pro vytipování optimální fáze růstu a koncentrace buněk pro provedení CRISPR/Cas, stanovení citlivosti na antibiotikum pro selekci mutantů a ověření přítomnosti cílového lokusu v kultuře Vischeria sp. C074. Tato část je dále rozšířená o analýzu genomového assembly a návrh primerů pro PCR.

Abstract:

The thesis deals with eustigmatophytes as model organisms in molecular biology, focusing on the strain Vischeria sp. C074, whose basic characteristics and biology (e.g. growth or sensitivity to antibiotics) are crucial for its subsequent use as a model eustigmatophyte. This bachelor thesis consists of two parts, theoretical and practical. The theoretical part focuses on the characteristic features of the algal group eustigmatophyte its classification including the history and present of this group, practical application focusing on the important representatives Nannochloropsis oceanica and Visheria. Furthermore, this work summarizes the methods of molecular biology used for genetic modification of organisms with specific examples of application on eustigmatophytes. The practical part focuses on obtaining the basic information that must be determined before modifying an organism. It includes the determination of the growth curve to identify the optimal growth phase and cell concentration for performing CRISPR/Cas, determination of antibiotic sensitivity for mutant selection and verification of the presence of the target locus in the culture of Vischeria sp. C074. This section is further expanded to include genome assembly analysis and primer design for PCR.
