Antibiotic resistance of methanogenic archaea in human microbiota

Abstract:

Vznik domény Archaea, na základě práce Woeseho a Foxe v roce 1977, způsobil revoluci v oblasti evoluční biologie. Zástupci této nové domény byli z počátku nalézáni převážně v prostředích s extrémními podmínkami. Až relativně v nedávné době, s rozvojem citlivějších technik detekce a kultivace těchto mikroorganismů, byly nalezeni zástupci, kteří jsou součástí mikrobioty různých živočichů včetně člověka. V současnosti lze konstatovat, že z hlediska posuzování možné patogenity, nebyly nalezeny informace poukazující na skutečnost, že by archea v lidské mikrobiotě působily jako patogeny. Nicméně se na patogenitě mohou symbioticky účastnit. Je nutno obecně konstatovat, že rozsah informací ohledně role archea v rámci symbiózy s bakteriemi během metabolických pochodů v GIT je poměrně omezený, rovněž tak jejich potenciál na rozvoj specifických onemocnění. Při léčení různých onemocnění se zástupci archea také dostávají do kontaktu s antibiotiky. Cílem testování bylo ověření rozsahu antibiotické resistence u vybraného zástupce metanogenních archea Methanobrevibacter smithii ke skupině vybraných antibiotik. V rámci experimentální části jednotlivé testy probíhaly za definovaných podmínek s využitím specifických metod detekce pro zhodnocení jejich růstu a životaschopnosti. Bylo ověřeno že Methanobrevibacter smithii je citlivý na chloramfenikol a zcela rezistentní vůči působení ampicilinu, kanamycinu i vankomycinu. Poslední jmenované antibiotikum způsobilo ve vyšších koncentracích dokonce nárůst biomasy oproti nižším koncentracím tohoto antibiotika během testování.

Abstract:

Establishing of a domain Archaea, on the basis of Woese and Fox study in the year 1977, started a revolution in the area evolutional biology. Representatives of this new domain were at first open up largely in environments with extreme conditions. Just in recent years, in relationship with development of new techniques and more sensitive detection and cultivation these microorganisms, were found species that are a part of different microbiomes of animals including humans. Presently it is possible to state, that in the light of current consideration there are no indications about their pathogenity, when archaea in human microbiota would react like pathogens, nevertheless on pathogenity they may symbioticly participate. Is necessary to express, that the range of information regarding the role of archaea in terms of symbiosis with bacteria during metabolic pathways in GIT is relatively limited, as well their potential contribution on development of specific of diseases. During medical treatment of various diseases some of archaeal species may get in contact with antibiotics. The aim of experiments was to check a range of antibiotic resistance related to a selected methanogenic archaeon typical for GIT, Methanobrevibacter smithii by a specific group of antibiotics. The experiments were proceeded according to defined plan and conditions with usage of specific kinds of detection devices for estimation archaeal growth and antibiotic resistance. It was observed that Methanobrevibacter smithii was susceptible to chloramphenicol and resistant to ampicillin, kanamycin and vancomycine. The last one had created an increased growth of biomass in higher concentrations compared to lower concentrations of this antibiotic during testing.
