The control of spindle assembly in mammalian oocytes and embryos

Abstract:

Dělicí vřeténko je jedním z nejdůležitějších nástrojů buňky zabezpečující zachycení, srovnání a rozdělení chromozomů v průběhu buněčného dělení. Je to samo sestavovací mikrotubulární struktura nevyhnutelná ke správné segregaci chromozomů. Přísná kontrola sestavení a udržení dělicího vřeténka je v mitóze velmi důležitá, aby se zabránilo chybám. V této práci jsme se soustředili na mechanizmy regulace délky dělicího vřeténka, konkrétně na regulační funkci velikosti buňky a poměru jádra a cytoplazmy. Jelikož se blastomery na začátku vývoje embrya dělí, aniž by buňky zvětšovaly objem, buněčné organely a struktury musí svoji velikost přizpůsobit velice rychle. V žabích i myších embryích byl popsán jev, kdy dělicí vřeténko dosahuje ve velmi velkých buňkách své maximální délky, tzv. horního limitu. V této práci jsme objem dvoubuněčných blastomer zdvojnásobili a ztrojnásobili pomocí elektrofúze. Dělicí vřeténko bylo v těchto zvětšených buňkách značně delší, což zpochybňuje teorii horního limitu délky vřeténka u myších embryí. Naše experimenty ukazují spíše na regulační funkci poměru jádra a cytoplazmy než velikosti buňky. Pomocí elektrofúze, enukleace a partenogenetické aktivace se nám podařilo vytvořit buňky, které měly nižší poměr jádra a cytoplazmy než intaktní embrya. U těchto buněk jsme zaznamenali výrazně delší dělicí vřeténko, a tak se domníváme, že v poměru jádra a cytoplazmy existuje rovnováha, která musí být pro správnou regulaci délky dělicího vřeténka v raném myším embryu zachována. Ve druhé části této práce jsme se zaměřili na raný vývoj embrya, především na charakterizaci délky jednotlivých buněčných cyklů, poruch v mitóze a aneuploidii. Údaje z reprodukčních klinik ukazují na značnou míru aneuploidie v lidských embryích. V našich experimentech na myších embryích jsme zaznamenali abnormality v mitóze typu vychýlených a opožděných chromozomů až u 15 % blastomer. Hranice mezi takto abnormálním chováním chromozomů v dělení a aneuploidií je velice tenká. V jedno-, dvou- a osmi- buněčných embryích jsme pozorovali aneuploidii u 5 % blastomer, převážně to byly ztráty nebo zisky jednoho až dvou chromozomů. U lidských embryí v stadiu osmi buněk je pozorován zcela jiný profil aneuploidie, embrya jsou často vysoce aneuploidní, tzv. mozaiková.

Abstract:

Spindle apparatus is one of the most important tools in the cell for the capturing, aligning and segregation of chromosomes during division. It is a microtubule based self-assembling structure important for the correct chromosome segregation. Tight regulation of the spindle assembly and maintenance is important to prevent errors in mitosis. In this work, we focused on regulation of spindle length in early mouse embryo, namely on regulatory function of the cell size and nucleus-to-cytoplasm ratio. Early embryos are the challenging system for the spindle size scaling because blastomeres divide several times without intervening growth for the first few days after fertilization. It has been suggested, based on studies on Xenopus and mouse embryos, that spindle reaches its upper length limit in large cells at the beginning of the development. In our study we increased the volume of 2-cell embryo blastomeres two- or three-folds using the electrofusion method. We observed a significant increase in the spindle length in these large cells, and this finding questions the proposed theory of an upper limit of the spindle length in a mouse embryo. Our experiments indicate the regulatory role of nucleus-to-cytoplasm balance, rather than the cell size. In our further experiments, we created the cells with lower nucleus-to-cytoplasm ratio than normal by electrofusion, enucleation and parthenogenetic activation and such cells possess considerably longer spindle. Our findings indicate that the nuclear to cytoplasmic ratio must be preserved for the correct regulation of the spindle length in early mouse embryo. In the second part of this work, we focused on the preimplantation embryo development, particularly the progression through each cell cycle, chromosome segregation abnormalities and aneuploidy rates. Data from IVF clinics show that human embryos are susceptible for high level of aneuploidy. We used live cell imaging to examine early mouse embryos, and we detected mitotic abnormalities, such as misaligned or lagging chromosomes. According to our observation, 5 % of blastomeres in 1-cell, 2-cell and 8-cell embryo were carried aneuploidy. Mostly, there were simple losses or gains of 1 or 2 chromosomes, in contrast to human embryo with massive mosaic aneuploidy in cleavage stage.
