Leishmania virulence: from haplotype network to functional genomics

Abstract:

The genus Leishmania is a species-rich member of the family Trypanosomatidae. These parasites cause a spectrum of diseases in humans, as well as in wild and domestic animals. Species of this genus are highly heterogenous and their phylogeographic pattern is rather complex. In this thesis, I reviewed geographic distribution of L. tropica haplotypes, the causative agent of cutaneous leishmaniasis in the Old World, and discussed genes involved in Leishmania virulence. The thesis contains four publications that I co-authored, all of them investigating biology of Leishmania spp. from the molecular perspective. The first one illustrates the global phylogeographic structure of L. tropica haplotypes and presents evidence supporting zoonotic transmission of this species that was previously considered to be anthroponotic. Two other publications characterize novel virulence factors of Leishmania (Oxygen-sensing adenylate cyclase (LmxM.28.0090) and dual specificity protein/lipid phosphatase (LmxM.22.0250)). Besides, these two publications demonstrate the power of CRISPR/Cas9 technology for effective gene editing in Leishmania genomes. The last publication addresses the frequency of putative recombination/gene conversion events and their patterns in L. donovani genomes. Advances in sequencing technologies, development of other modern molecular biological techniques (e.g., CRISPR/Cas9), and their application in Leishmania spp. allowed us to extend our knowledge in phylogeography, genetics, and virulence of Leishmania.

Abstract:

Rod Leishmania je druhově bohatou linií čeledi Trypanosomatidae. Tito parazité způsobují spektrum nemocí jak u člověka, tak i u divokých a domácích zvířat. Druhy tohoto rodu jsou velice heterogenní a jejich fylogeografie je poměrně komplexní. V této práci shrnuji geografické rozšíření haplotypů patřících do druhu L. tropica, původce kutánní leishmaniózy ve Starém světě, a diskutuji geny ovlivňující virulenci rodu Leishmania. Práce obsahuje čtyři publikace, na kterých jsem se podílela jako spoluautor, všechny se zabývají biologií rodu Leishmania z molekulární perspektivy. První ilustruje globální fylogeografickou strukturu haplotypů L. tropica a předkládá důkazy podporující zoonotický přenos tohoto druhu, který byl dosud považován za antroponózu. Další dvě publikace se zabývají charakterizací nových virulenčních faktorů u leishmanií (Oxygen-sensing adenylate cyclase LmxM.28.0090; dual specificity protein/lipid phosphatase LmxM.22.0250). Navíc obě tyto publikace ukazují sílu CRISPR/Cas9 technologie pro efektivní genový editing leishmanií. Poslední publikace se zaměřuje na frekvenci pravděpodobných událostí rekombinace/genové konverze a jejich distribuce v genomech L. donovani. Pokroky v sekvenačních technologiích, vznik dalších moderních molekulárně-biologických technik (např. CRISPR/Cas9) a jejich použití při výzkumu leishmanií nám umožnily rozšířit znalosti o fylogeografii, genetice a virulenci rodu Leishmania.
