Abstract:

\recke{alpha}This dissertation study is focused on the development of the liposomal formulations of hydrophobic anticancer drugs. The first part of the study deals with the preparation of a stable liposomal formulation capable to encapsulate a high content of PTX. Various lipid compositions were screened in order to find the optimal composition for the preparation of physically and chemically stable liposomes as well as to develop the liposomes with a high encapsulation capacity. The next part of the study was focused on the development of suitable liposomal formulation for pro-apoptotic analogues of VE such as alpha-TOS and alpha-TAM. Unlike alpha-TAM, alpha-TOS is commercially available. The last part of the study investigates a potential toxic effect of VE analogues. The toxicity of free and liposomal formulations of VE analogues was tested in vitro and in vivo. The anticancer effect of the liposomal formulations of both alpha-TAM and alpha-TOS was evaluated in the mice with implanted hollow fibres and in the transgenic mice bearing spontaneous breast carcinomas.

Abstract:

Tato disertační práce se zabývá vývojem liposomálních preparátů hydrofobních antikancerogenních látek.V první části se pozornost soustředila na přípravu stabilního liposomálního preparátu, který byl schopen enkapsulovat vysoký obsah paclitaxelu. Různé lipidní kompozice byly testovány za účelem najít vhodné optimální složení liposomů a připravit fyzikálně a chemicky stabilní liposomy s vysokou enkapsulační kapacitou.Další část práce se zabývala vývojem vhodných liposomálních preparátů proapoptotických analogů vitaminu E, a to pro alfa-tokoferyl sukcinát (alfa-TOS) a alfa-tokoferyl amidomalát (alfa-TAM). Poslední část se věnovala studiu toxicity analogů vitaminu E. Toxicita volných a liposomálních forem analogů vitaminu E byla testována in vitro a in vivo. Antikancerogenní účinek obou liposomálních preparátů alfa-TOSu a alfa-TAMu byl ověřen na myších nádorových modelech (implantáty "hollow fibre" a transgenní myši se spontánním prsním karcinomem).
