Genetic structure of freshwater shrimp (Gammarus fossarum) populaton in stream headwater zones

Abstract:

Hlavním cílem této práce bylo popsat populačně-genetickou strukturu blešivce potočního (Gammarus fossarum) v pramenných úsecích toků a ověřit, zda je závislá na struktuře říční sítě. Blešivec potoční je zástupcem permanentní fauny, obývající prameny, pramenné stružky a ritrální úseky toků. Lze proto předpokládat, že jeho schopnost aktivní disperze přes terestrické prostředí bude do značné míry omezena. Celkem bylo genotypizováno 664 jedinců z 24 populací na pěti polymorfních mikrosatelitových lokusech. Vzorky pocházely z pramenů a pramenných stružek z oblasti moravsko-slovenského pomezí, přičemž 3 populace náležely do povodí Odry, 10 do povodí Moravy a 11 do povodí Váhu. Populace G. fossarum byly geneticky značně diferencované (FST = 0,279, p < 0,001) a s jejich rostoucí vzdáleností se mezi nimi zvyšovala i vzdálenost genetická (dle modelu „isolation by distance“). Na malých prostorových škálách (cca 200 m) však byly geneticky téměř uniformní. Bylo zjištěno, že rozvodí mezi studovanými povodími nemusí být nepřekonatelnou bariérou zabraňující genovému toku mezi lokálními populacemi tohoto druhu. Blešivci tak zjevně jsou schopni překonat terestrické prostředí. Nebyl nalezen významný vztah mezi genetickou diverzitou populací a nadmořskou výškou ani pH. Dle předpokladu korelovala s genetickou diverzitou konduktivita (jako proxy proměnná vápnitosti lokality), avšak poměrně slabě.

Abstract:

The main aim of this study was to describe the population genetic structure of the freshwater shrimp (Gammarus fossarum) in springs in order to test if it depends on the hierarchical river structure. G. fossarum is representative of permanent fauna living in springs, springbrooks and rhitral streams, so it can be assumed that its capability of active dispersion through the terrestrial environment will be considerably limited. In total, 664 individuals from 24 populations were genotyped on five polymorphic microsatellite loci. Samples originated from springs and springbrooks in the Outer Western Carpathian Mts. 3 populations were located in the Oder basin, 10 in the Morava drainage system and 11 in the Váh basin. G. fossarum showed a strong genetic differentiation (FST = 0,279, p < 0,001) and a clear pattern of isolation by distance. However, populations were genetically almost identical at small spatial scales (approximately 200 m). It was found out that a major watershed between drainage basins may not be an insuperable barrier to gene flow among local populations of this species. This indicates that G. fossarum might be capable of dispersing overland in headwater regions. There was no significant correlation between altitude or pH and genetic diversity within populations. Only weak correlation was observed between genetic diversity and water conductivity as a proxy of calcium concentration at the locality.
