Oxygen transfer investigation in Acidithiobacillus ferrooxidans cultures

Abstract:

Práce přináší konkrétní data charakterizující limitaci biooxidace Fe(II) a S kyslíkem. Tato zjištěná data sloužila dále k určení specifických podmínek pro stanovení minimální hodnoty objemového koeficientu přestupu kyslíku kLa, jež v následujícím pokusu zajistil kyslíkem nelimitovanou oxidaci Fe(II) a S. Rozdíly mezi hodnotami Michaelisovy konstanty KM pro kyslík a hodnotami kLa pro kulturu A. ferrooxidans oxidující Fe(II) a S jsou statisticky významné. Kultura oxidující síru má nižší požadavky na kyslík než kultura oxidující Fe(II). Sirná kultura ve fermentoru má nižší požadavky na kyslík než sirná kultura na třepačce. Pro svoji univerzálnost mohou hodnoty kLa sloužit k modelování podmínek aerace v bioreaktorech.

Abstract:

This thesis provides specific data describing oxygen limitation in biooxidation of Fe(II) and elemental sulfur. These data were also used for prediction of the specific conditions that determine the minimum value of the volumetric oxygen transfer coefficient kLa. In the following experiment, this coefficient secured the biooxidation of Fe(II) and elemental sulphur not limited by oxygen consumption. Differences between the values of Michaelis constant KM for oxygen and values of kLa for bacterial culture A. ferrooxidans oxidizing Fe(II) and elemental sulfur, are statistically significant. Sulfur oxidizing culture has lower oxygen demands than a culture oxidizing Fe(II). Sulfur oxidizing culture in the fermenter has lower oxygen demands than sulfur oxidizing culture on a shaker. Because of its universality, kLa values may be used for modelling the conditions of aeration in bioreactors.
