Molecular identification and variation of amphibians in the central Congo Basin

Abstract:

Biodiverzita střední části Konžské pánve je bohatá, avšak velmi málo prozkoumaná nejen s ohledem na herpetofaunu, ale i další skupiny drobných obratlovců. Jedním z důvodů je, že velkou část tohoto území pokrývají nížinné deštné lesy, které jsou velmi špatně přístupné. I přesto zde dochází k nelegálnímu těžbě dřeva, což vede ke ztrátě biotopů, které jsou pro tyto živočichy nepostradatelné. Cílem této práce byla základní druhová identifikace vzorků pomocí molekulárních metod a srovnání získaných sekvencí s online databází GenBank. Bylo identifikováno 56 druhů obojživelníků, z nichž některé nebyli dosud popsány a představují tedy taxony pro vědu nové. Byla také zhodnocena genetická variabilita jednotlivých druhů mezi sledovanými oblastmi (Domaine de chasse de Rubi-Télé, Réserve de biosphère de Yangambi a Reserve neturelle de bonobo de Kokolopori). Ta byla následně zhodnocena s ohledem na bariéru řeky Kongo. Z těchto výsledků vyplývá, že popu-lace některých druhů jsou řekou Kongo přímo odděleny a vykazují tedy vysokou variabilitu, a naopak jiné druhy (striktně akvatické a synantropní) nevykazují téměř žádnou variabilitu mezi populacemi řekou oddělenými. Tyto výsledky byly vyhodnoceny v kontextu literární rešerše o biogeografii a diverzitě obojživelníků a dalších skupin organismů střední Afriky.

Abstract:

The biodiversity of the central part of the Congo Basin is rich, but very little explored not only with regard to the herpetofauna, but also other groups of small vertebrates. One of the reasons is that a large part of this area is covered by lowland rainforests, which are very difficult to access. Even so, illegal logging takes place here, which leads to the loss of habitats that are indispensable for these animals. The aim of this work was basic species identification of samples using molecular methods and comparison of obtained sequences with the online GenBank database. 56 species of amphibians have been identified, some of which have not yet been described and thus represent taxa new to science. The genetic variation of individual species between the monitored areas (Domaine de chasse de Rubi-Télé, Réserve de biosphère de Yangambi and Reserve neturelle de bonobo de Kokolopori) was also evaluated. This was subsequently evaluated with regard to the Congo River barrier. These results show that populations of some species are directly separated by the Congo River and therefore show high variability, while other species (strictly aquatic and synanthropic) show almost no variation between populations separated by the river. These results were evaluated in the context of a literature review on the biogeography and diversity of amphibians and other groups of organisms of Central Africa.
