NMR spectroscopy of medically and pharmaceutically interesting nitrogen heterocycles

Abstract:

Pomocí nukleární magnetické rezonance (NMR) byly studovány dvě skupiny dusíkatých heterocyklů - protoberberinové alkaloidy a purinové deriváty. Ze skupiny protoberberinových alkaloidů byly vybrány dva alkaloidy - - palmatin a berberin. Oba vykazují značnou biologickou aktivitu (např. cytotoxické účinky), vytvářejí také nekovalentní komplexy s DNA. Kvarterní protoberberinové alkaloidy jsou díky polární vazbě náchylné k nukleofilnímu ataku za tvorby příslušných aduktů. Studium těchto aduktů (izolačních artefaktů protoberberinových alkaloidů) a jejich NMR charakterizace byly předmětem této části dizertační práce. Purinové deriváty vykazují např. antibakteriální či protinádorové účinky. Mohou se vyskytovat v několika tautomerních formách. Vzhledem k rychlé chemické výměně mezi tautomery vidíme často v NMR spektru za laboratorní teploty pouze signály, které odpovídají časově zprůměrovanému příspěvku jednotlivých tautomerů. Měření NMR spekter za nízkých teplot pak v některých případech umožňuje separaci signálů jednotlivých tautomerů díky zpomalení vzájemné chemické výměny. Pomocí NMR spektroskopie za nízkých teplot byla zkoumána série různě substituovaných purinových derivátů. Charakteristické hodnoty NMR chemických posunů a interakčních konstant poskytují informaci o tautomerech přítomných v roztoku daného derivátu. Znalost tautomerních rovnováh umožňuje lepší vhled do interakcí purinových derivátů s biologickými cíli.

Abstract:

In this project, two groups of nitrogen heterocycles were investigated by NMR spectroscopy – protoberberine alkaloids and purine derivatives. Quaternary protoberberine alkaloids (QPA), naturally occurring in Berberidaceae, Fumariaceae and other plant families, are interesting for their chemistry and biological activities – some of them display antimalarial, cytotoxic and other effects and their interaction with nucleic acids has been frequently studied. The alkaloids are sensitive towards nucleophilic reagents and the free bases formed may adopt different structures. Studying the free bases of several protoberberine alkaloids and characterization of QPA isolation artifacts were some of the goals of this work. Purine derivatives display a various range of biological activities, for example antiviral and antitumor. They may exist in several tautomeric forms. The fast chemical exchange between tautomers at laboratory temperature causes the broadening of the NMR signals, while at low temperatures the chemical exchange is slowed down and signals corresponding to individual tautomers are often sharpened. The characteristic values of NMR chemical shifts and spin-spin coupling constants provide the important information about the protonated sites of the present tautomers. Knowledge of the tautomeric equilibria is important for better understanding the interaction of purine derivatives with biological targets.
