Hyaluronic acid

Abstract:

Práce se zabývá hyaluronovou kyselinou, která je pro své specifické a výjimečné vlastnosti využívána v medicíně i v kosmetickém průmyslu už 80 let. Prezentuje její výskyt, chemickou strukturu a využití v praxi. Hyaluronová kyselina je lineární polysacharid, skládá se z opakujících se jednotek disacharidu tvořeného N-acetyl-D-glukosaminem a D-glukuronovou kyselinou. Dospělé lidské tělo obsahuje až 15 g této kyseliny, která je každý den obnovována a po rozštěpení se hyaluronová kyselina transportuje lymfatickým systémem do krve, dále pak degradovaný hyaluronan je vyloučen játry do ledvin a následně a pryč z těla. Díky svý unikátním viskoelastickým vlastnostem a své vysoké molekulové hmotnosti se hyaluronová kyselina využívá v kosmetologii, oftalmologii, revmatologii, otolaryngologii, dermatologii, plastické chirurgii a při hojení ran. Popsán je zde i chondroitin sulfát, který se řadí mezi glukosaminglykany, nachází se v chrupavkách a dále v pojivových tkáních. Používá se například při léčbě revmatoidních onemocnění. Průmyslově se získává extrakcí z hovězích a vepřových chrupavek. Praktická část této práce se věnuje stanovení hyaluronové kyseliny vysokoúčinou kapalinovou chromatografií (HPLC). Stanovovaly se standardy nasycených a nenasycených oligomerů hyaluronové kyseliny (dimer-dekamer) a dále také hyaluronan štěpený enzymaticky hydrolasou a lyasou na oligomery. Povedlo se připravit ze standardů, kalibrační roztoky, ty byly proměřeny HPLC metodou a byly vytvořeny chromatogramy jednotlivých oligomerů. Standardy poskytnuté firmou Contipro Pharma a.s. hyaluronanu štěpené hydrolasou a lysaou, byly úspěšné, ale enzymaticky štěpené reálné (nativní) vzorky prasečích oocytů, se nepodařilo stanovit, pro jejich vysoký podíl nežádoucích složek.

Abstract:

Bachelor thesis deals with hyaluronic acid which is used in medicine and in the cosmetics for its specific and unique properties for over 80 years. The work presents its occurrence, chemical structure and it's use in practice. Hyaluronic acid is a linear polysaccharide and it consists of repeating units of disaccharide consisting of N-acetyl-D-glucosamine and D-glucuronic acid. The adult human body contains up to 15 g of this acid which is renewed every day, and after digestion hyaluronic acid is transported by the lymphatic system to the blood and then degraded hyaluronan is eliminated in the kidneys and liver and consequently away from the body. Due to it's unique viscoelastic properties and it's high molecular weight, hyaluronic acid is used in cosmetology, ophthalmology, rheumatology, otolaryngology, dermatology, plastic surgery and wound healing. The work also describes chondroitin sulfate which amogs to the glycosaminoglycans and it is located in cartilages and connective tissues. In the medicine CS is used for example in the treatment of rheumatic diseases. It is industrially obtained by extraction from beef and pork cartilage. The practical part of this thesis deals with the determination of hyaluronic acid by high performance liquid chromatography (HPLC). There are determined standards of saturated and unsaturated oligomers of hyaluronic acid (dimer-decamer) and also cleaved hyaluronate lyase enzyme and a hydrolase in oligomers. I managed to prepare calibration solutions from the standards. Solutions were measured by HPLC method and chromatograms of individual oligomers were created. Company Contipro Pharma a.s. has provided standards which were cleaved by hydrolase and lyase and they were successful. On the other side I did not determine native samples of porcine oocytes, which were enzymatically cleaved, because they contained a high proportion of undesirable components.
