Substances affecting the motility and chemotaxis of human sperm

Abstract:

Proces fertilizace je klíčovou událostí pro zrození nového organismu. Než se tak stane a dojde k fúzi obou membrán pohlavních buněk, musí nejprve tyto gamety mezi sebou intenzivně komunikovat. Chemotaxe spermií se zdá být rozšířeným mechanismem, který navádí spermie k vajíčku, a to jak v případě vnějšího, tak vnitřního oplození. Tento proces umožňuje spermii vnímat a pohybovat se směrem k vzestupnému gradientu chemických molekul (chemoatraktantů), a tak dorazit k vajíčku. Specifické receptory exprimované na povrchu spermie vnímají v prostředí chemické signály a spouští rozdílné signální dráhy, které následně ovlivní pohyblivost a regulují správnou funkci spermie. Přestože vliv různých látek na chemotaxi a pohyblivost spermií může mít velký význam pro plodnost a asistovanou reprodukci, je v literatuře věnována na toto téma relativně malá pozornost. Cílem diplomové práce bylo vybrat rutinně použitelnou metodu studia chemotaxe a pohyblivosti lidských spermií a otestovat látky, které mohou ovlivňovat tyto jevy v pozitivním i negativním smyslu. Metodou Choice assay bylo zjištěno, že nejvíce spermií je přitahováno kofeinem, folikulární tekutinou a teofylinem. Naopak chemikálie, které nejméně vykazovaly vliv na pohlavní buňky, byly kinetin a nobiletin.

Abstract:

The process of fertilization is a key event for the birth of a new organism. Before this happens and the fusion of the two sex cell membranes takes place, these gametes must first communicate intensively with each other. Sperm chemotaxis appears to be a widespread mechanism that guides sperm to the ovum in both external and internal fertilization. This process allows the sperm to perceive and move towards the increasing gradient of chemical molecules (chemoattractants) and thus reach the ovum. Specific receptors expressed on the surface of the sperm perceive chemical signals in the environment and trigger different signalling pathways that subsequently affect the motility and regulate the proper functioning of the sperm. Although the effect of various substances on sperm chemotaxis and motility may be of great importance for fertility and assisted reproduction, relatively little attention is paid to this topic in the literature. The aim of this master’s thesis was to select a routinely applicable method of studying chemotaxis and motility of human spermatozoa and to test substances that may influence these phenomena in both a positive and negative sense. Using the Choice assay method, it was found that most sperm are attracted to caffeine, follicular fluid, and theophylline. Conversely, the chemicals that showed the least effect on male gametes were kinetin and nobiletin
