Preparation and characterization of lectins from bacterium Photorhabdus luminescens

Abstract:

Lectins belong to the group of proteins that specifically and reversibly bind carbohydrates. One of the biological consequences of this interaction is a molecular recognition in host-pathogen interactions. Bacterial lectins may function as virulent factors that enable bacteria to bind surface glycans on the host cells and initiate colonization and host infection. This is one of the reasons why some of the bacterial lectins are currently viewed as potential targets for anti-adhesion therapy. The Master's thesis is focused on the structural and functional characterization of lectins from Photorhabdus luminescens bacteria and builds on a previous work published in the Bachelor's thesis Cloning of lectins from Photorhabdus spp. A novel PLL lectin from P. luminescens has been characterized recently. After bioinformatics analysis, we found four hypothetical proteins with high sequence homologies to the PLL lectin. Their functions and properties have yet to be characterized. The thesis deals with two proteins. The first part is focused on the PLU34 lectin characterization that represents the core of the thesis. The second part is focused on the PLU35 lectin. The work is based on the protein biochemistry techniques including protein purification, crystallization, determination of protein oligomerization state and characterization of the binding specificities using biophysical methods such as analytical ultracentrifugation, isothermal titration calorimetry and surface plasmon resonance.

Abstract:

Lektiny patří mezi skupinu proteinů, které dokáží reverzibilně a specificky interagovat se sacharidy. Jedním z biologických následků této interakce je molekulární rozpoznání v rámci hostitelsko-patogenních interakcí. Bakteriální lektiny mohou působit jako virulentní faktory, které umožňují vazbu na povrchové sacharidy hostitelských buněk a tím iniciovat kolonizaci a infekci hostitele. Proto jsou v současné době některé bakteriální lektiny vnímány jako potenciální cíle nových antiadhesivních léčiv. Diplomová práce se zabývá strukturně-funkční charakterizací lektinů z bakterie Photorhabdus luminescens, přičemž navazuje na bakalářskou práci Klonování lektinů z Photorhabdus spp. Nedávno byl charakterizován nový lektin PLL z P. luminescens. Po bioinformatické analýze byly objeveny čtyři hypotetické proteiny s vysokou sekvenční homologií k lektinu PLL, jejichž vlastnosti a funkce jsou zatím nejasné. V práci se věnujeme dvěma proteinům. První část je věnována charakterizaci lektinu PLU34, která tvoří jádro práce. Druhá část je věnována lektinu PLU35. Práce je založena na metodách proteinové biochemie zahrnujících proteinovou purifikaci, krystalizaci, charakterizaci oligomerného stavu a vazebné specifity daných lektinů s využitím biofyzikálních metod jako je analytická ultracentrifugace, isotermální titrační kalorimetrie a rezonance povrchového plasmonu.
