Detection of herbicide resistant weeds using techniques of molecular biology

Abstract:

Tato bakalářská práce se zabývá charakteristikou plevelů spolu se způsoby jejich regulace, zejména pomocí herbicidů, a molekulární detekcí rezistentních jedinců. V praktické části této práce byla provedena molekulární detekce rezistentních plevelů. Celkem byly sledovány tři geny. Dva z těchto genů, ALS a ACC2 byly sledovány u ježatky kuří nohy (Echinochloa crus-galli (L.) P. Beauv.), u kterých substitucí určitých aminokyselin může dojít ke vzniku rezistence vůči herbicidům inhibujícím acetolaktátsynthasu nebo acetylkoenzymAkarboxylasu. Dva z těchto genů, ALS a EPSPS1 u turanky kanadské (Conyza canadensis L. Cronquist), u kterých substitucí určitých aminokyselin může dojít ke vzniku rezistence vůči herbicidům inhibujícím acetolaktátsynthasu nebo 5-enolpyruvylšikimát-3-fosfátsynthasu. Mutace, souvisejících přímo se vznikem rezistence rostlin vůči herbicidům inhibujícím ALS, ACC nebo EPSPS, nebyly detekovány.

Abstract:

This bachelor thesis is concerned with the characterization of weeds together with methods of their regulation using various herbicides; and with molecular detection of resistant specimens. In the practical part of this thesis, a molecular detection of resistant weeds was performed. In total, three genes were observed. Two of these genes, ALS and ACC2 from barnyard grass (Echinochloa crus-galli (L.) P. Beauv.), might undergo the substitution of specific amino acids which can cause development of resistance to herbicides that inhibit acetolactate synthase or acetyl coenzyme A carboxylase. In other two genes, ALS and EPSPS1 from horseweed (Conyza canadensis L. Cronquist), the substitution of specific amino acids can cause the development of resistance to herbicides that inhibit acetolactate synthase or 5-enolpyruvoylshikimate-3-phosphate synthase. Mutations related to development of resistance in plants, which might be connected with herbicides inhibiting ALS, ACC or EPSPS, were not detected.
