-Use of stable isotopes to evaluate the response of different plant species and its dependence on climatic conditions

Abstract:

Interakce rostlin s růstovým prostředím se odráží v izotopovém složení jejich orgánů a pletiv. V rámci disertační práce jsem studovala poměry stabilních izotopů uhlíku (13C/12C), kyslíku (18O/16O) a dusíku (15N/14N) v několika druzích rostlin (jednoleté polní plodiny, dlouhověké dřeviny). Naše studie prokázaly, že izotopové složení celulózy extrahované z jednotlivých letokruhů dřevin citlivě odráží meziroční změny hydroklimatických podmínek. Zatímco poměr 13C/12C citlivě reaguje zejména na změny vláhového režimu, poměr 18O/16O odráží změny v teplotním režimu dané lokality. K pochopení změn fyziologických procesů spojených s cykly vody a uhlíku v lesních ekosystémech jsem na základě izotopového poměru 13C/12C v letokruzích dubů rekonstruovala efektivitu využití vody (WUE) v gradientu nadmořských výšek. Pomocí stanovení izotopového složení zrn pšenice ozimé bylo prokázáno, že i krátkodobé (několikadenní) působení kombinace sucha a zvýšené teploty může vést k dlouhodobému zvýšení WUE i po konci stresových podmínek. Pro potřeby archeologických rekonstrukcí jsem dále studovala, jak se izotopový signál zachovává ve zuhelnatělých archeologických nálezech zrn. Izotopové složení experimentálně zuhelnatělých zrn prokázalo velmi dobré zachování izotopových poměrů uhlíku a částečně i dusíku, a lze je proto aplikovat v rámci archeologických výzkumů k porozumění dávných růstových podmínek a způsobu hospodaření.

Abstract:

The interaction of plants with their growth environment is reflected in the isotopic composition of their organs and tissues. As part of my dissertation, I studied the ratios of stable isotopes of carbon (13C/12C), oxygen (18O/16O) and nitrogen (15N/14N) in several types of plants (annual field crops, long-lived trees). Our studies showed that the isotopic composition of cellulose extracted from individual tree rings sensitively reflects interannual changes in hydroclimatic conditions. While the ratio 13C/12C reacts particularly sensitively to changes in the water regime, the ratio 18O/16O reflects changes in the temperature regime of the given location. Compared to the traditionally used tree ring width, higher sensitivity and higher correlation coefficients with hydroclimatic parameters were demonstrated for isotopic ratios. To understand the changes in physiological processes associated with water and carbon cycles in forest ecosystems, I reconstructed the water use efficiency (WUE) in an altitudinal gradient based on the 13C/12C isotopic ratio in oak tree rings. Using the determination of the isotopic composition of winter wheat grains, it was demonstrated that even short-term (several days) exposure to a combination of drought and elevated temperature can lead to a long-term increase in WUE even after the end of the stressful conditions. For the needs of archaeological reconstructions, I further studied how the isotopic signal is preserved in charred archaeological finds of grains. The isotopic composition of the experimentally charred grains had shown a very good preservation of the isotopic ratios of carbon and partly also of nitrogen, and can therefore be used in archaeological research to understand the ancient conditions of growth and the way of farming.
