Preparation of microparticles by cryogenic grinding nanofiber layers of hyaluronic acid derivates

Abstract:

V dnešní době jsou nanovlákna v popředí zájmu vědy a testují se jejich možné aplikace v různých odvětvích vědy a průmyslu. Díky svým jedinečným vlastnostem a významné podobnosti s extracelulární tekutinou nacházejí využití na poli tkáňového inženýrství. Mezi vhodné materiály na přípravu nanovláken patří především polymery a stále více se dává přednost polymerům přirozeně se vyskytujícím. Zájem se v poslední době soustředí na kyselinu hyaluronovou a její deriváty. Tato diplomová práce se zabývá přípravou mikročástic pomocí kryogenního mletí nanovlákenných vrstev dvou derivátů kyseliny hyaluronové. Nanovrstvy jsou připravovány elektrospinningem pomocí přístroje 4SPIN. Práce se soustředí na vliv podmínek procesu přípravy mikročástic a vzájemně porovnává vlastnosti obou derivátů. U takto připravených mikročástic se sleduje především jejich velikost, distribuce velikosti a cytotoxicita. Bylo zjištěno, že oba deriváty se liší svou strukturou nanovláken, což může mít vliv na velikost částic a jejich cytotoxicitu. Různé podmínky přípravy mikročástic hrají roli v distribuci částic, ale nikoliv v jejich cytotoxicitě. Oba deriváty jsou využitelné na poli biomedicíny. Podle konkrétní budoucí aplikace je potřebné je ale dále vhodně testovat.

Abstract:

Recently, nanofibers are of utmost interest in the field of science and potential nanofiber applications are being discovered and tested in various fields of science and industry. Nanofibers are widely used in tissue engineering mainly due to their extraordinary capabilities and structural similarities with the extracellular matrix. Polymers represent one of the most suitable materials for nanofabrication. Special emphasis is put on naturally occurring polymers such as hyaluronic acid and its derivatives. This diploma thesis deals with fabrication of microparticles by cryogenic grinding of nanofiber layers of two distinct hyaluronic acid derivatives. Nanofiber layers are produced by electrospinning with use of 4SPIN. Thesis is focused on characterization of microparticles of hyaluronic acid derivatives fabricated under different conditions of cryogenic grinding. We evaluated size of microparticles and its distribution, and cytotoxicity of microparticles. Both derivatives have had significantly different structure of nanofibers which in turn can have impact on cytotoxicity and size of fabricated microparticles. Different conditions tested in fabrication of microparticles affected distribution of microparticle sizes, but not their cytotoxicity. Both derivatives are potentially applicable in the field of biomedicine. However, further specialized tests should be conducted according to intended use of microparticles.
