Characterization of catalytic domains of selected carbonic anhydrases and interactions with their inhibitors

Abstract:

Karbonické anhydrázy jsou metaloenzymy dobře známé v celé přírodní říši. Hlavní úlohou rodiny α-karbonických anhydráz je katalýza fyziologicky nejdůležitějšího pufrovacího systému v lidském těle – hydrogenuhličitanového pufru. V lidském organismu se vyskytuje patnáct isozymů, které vykazují vysokou míru strukturní podobnosti. Karbonické anhydrázy jsou také spojovány s různými nemocemi, ale pouze isozymy II, IX a XII se spojují s rakovinou. Karbonická anhydráza IX je zajímavým cílem výzkumu vzhledem k její nadměrné expresi u agresivních typů rakoviny s expanzivními metastázemi a špatnou prognózou pro pacienta. Inhibitory karbonických anhydráz jsou široce používány jako terapeutika, přestože často vykazují neselektivní inhibici. Tato práce je zaměřena na přípravu a biofyzikální popis karbonických anhydráz souvisejících s rakovinou. Enzymy připravené v dostatečném množství a kvalitě byly dále testovány s novými inhibitory za účelem stanovení vazebných afinit a selektivity. Přestože všechny testované sloučeniny byly navrženy jako selektivní inhibitory karbonické anhydrázy IX, většina z nich vykazovala preferenční inhibici karbonické anhydrázy II. U jedné sloučeniny však bylo dosaženo selektivní inhibice karbonické anhydrázy IX.

Abstract:

Carbonic anhydrases are metalloenzymes well-known throughout the whole nature kingdom. The main role of the α-carbonic anhydrases family is the catalysis of the physiologically most important buffer system in the human body – the bicarbonate buffer. There are fifteen isozymes of these influential enzymes in humans which exhibit a high degree of structural similarity. Carbonic anhydrases are associated with various diseases but only isozymes II, IX, and XII are known to be related to cancer. Carbonic anhydrase IX is an interesting research target due to its overexpression in aggressive types of cancer with expansive metastasis. Carbonic anhydrase inhibitors are widely used as therapeutics although they often show non-selective inhibition and thus inhibit off-target carbonic anhydrases. This thesis is focused on the preparation and biophysical description of catalytic domains of cancer-related carbonic anhydrases. Enzymes prepared in sufficient quantity and quality were further tested with new potential carbonic anhydrase inhibitors to determine their binding affinities and selectivity. Although all tested compounds were designed as selective inhibitors of carbonic anhydrase IX, most of them showed a preferential inhibition of carbonic anhydrase II. However, one compound has been shown to inhibit carbonic anhydrase IX with a considerable degree of selectivity.
