Abstract:

Prase je historicky řazeno do skupiny živočichů, kteří tvoří své B lymfocyty v primárních lymfatických orgánech zadního střeva (tzv. GALT skupina). Cílem diplomové práce bylo ověřit, zda ileální Peyerské pláty prasat, které jsou předpokládaným místem vývoje B buněk u tohoto druhu, splňuji kritéria primárních lymfatických orgánů. Práce se konkrétně zaměřila na antigen nezávislý vývoj B buněk v ileálních Peyerských plátech. Vliv vnějších antigenů byl zkoumaný srovnáním lymfocytárních populací izolovaných z různých částí tenkého střeva konvenčních a bezmikrobních selat. Průtoková cytometrie fluorescenčně značených lymfocytů ukázala velké rozdíly mezi těmito experimentálními skupinami. Konkrétně šlo o zvýšení počtu B lymfocytů v ileu konvenčních zvířat v porovnání se zvířaty bezmikrobními. Toto zvýšení se dále projevilo relativním snížením všech ostatních základních lymfocytárních populací (αβ T lymfocytů, γδ T lymfocytů a NK buněk) v ileu konvenčních selat v porovnání se selaty bezmikrobními. Mikrobiální kolonizace také vedla k indukci IgA produkujících B lymfocytů a ke zvýšenému zastoupení efektorových stadií αβ T a γδ T lymfocytů v ileu. Tyto buňky jsou typické pro sekundární lymfatické orgány. Tyto a další nálezy ukazují, že ileální Peyerské pláty nejsou s největším pravděpodobností primární lymfatické orgány a prase tedy nelze nadále řadit do GALT skupiny podle vývoje B buněk.

Abstract:

Swine are speculated to belong to a group of animals that develop their B cells in the primary lymphoid organs located in the hindgut (so called the GALT group). The aim of this thesis was to elucidate whether porcine ileal Peyer's patches as the anticipated place of B cell development in swine meet the criteria of the primary lymphoid organ. The study is focused on issue of antigen-independent development of B cells in the ileal Peyer's patches. The effect of external antigen stimulation was determined by comparison of lymphocyte populations isolated from different parts of a small intestine of conventional and germ-free piglets. Flow cytometric analysis of fluorescently labeled lymphocytes showed the significant differences between these two experimental groups. Specifically, there was an increase in the number of B cells in ileum of conventional animals compared to germ-free animals. This increase in B cells resulted in a relative reduction of all the other basic lymphocyte subsets (αβ T cells , γδ T cells and NK cells) in ileum of conventional piglets compared to germ-free piglets. Microbial colonization also caused the induction of IgA-producing B cells and an increased representation of effector αβ and γδ T cells in the ileum. These changes are typical for a secondary lymphoid organ. The result of this study and further findings suggest that the development of B cells in ileal Peyer's patches is antigen-dependent. Therefore, ileal Peyer's patches are probably not a primary lymphoid organ in pigs and swine should not be classified as a member of the GALT group.
