Distribuce acetylovaného histonu H3 na lysinu 56 v průběhu buněčného cyklu nádorových a embryonálních kmenových buněk

Distribution of histone H3 acetylation on lysin 56 during cell cycle of malignant and stem cells

Abstract:

Acetylace histonu H3 na lysinu 56 je netradiční posttranslační modifikací histonu. Lysin 56 se nachází v globulární části histonu a jeho acetylace ovlivňuje stabilitu struktury samotného proteinu. Předpokládá se, že H3K56ac se účastní regulace regulace replikace, transkripce a oprav DNA v buněčném jádře. Nicméně, její role v buněčném cyklu není plně známa. Ve třech rozdílných typech buněčných linií (HeLa, HL-60 a hESC) s rozdílnou jadernou morfologií jsme sledovali zastoupení a distribuci H3K56ac během buněčného cyklu. H3K56ac je v jádře koncentrován do několika stovek ohnisek. V buněčném jádře pozorujeme ohniskovou distribuci H3K56ac. U nádorových linií se zvedá množství H3K56ac ohnisek v S a G2 fázi buněčného cyklu, zatímco u hESC linie je hladina stabilní. Přestože se u nádorových linií zvedá hladina samotného acetylovaného proteinu, poměr H3K56ac:H3 se v průběhu buněčného cyklu nemění. Zvýšená hladina H3 je tedy u nádorových buněk doprovázena také zvýšenou hladinou H3K56ac. Naše výsledky ukazují dynamickou distribuci H3K56ac v průběhu buněčného cyklu.

Abstract:

Histone H3 acetylation at lysine 56 is discrepant posttranslational histone modification. Lysine 56 is located in globular histone core and his acetylation influences the stability of protein structure itself. It´s supposed, that H3K56ac participates in regulation of replication, transcription and DNA reparation in cell nuclei. Nevertheless, the role of H3K56ac in a cell cycle hasn´t been fully elucidated, yet. The levels and distribution of H3K56ac during cell cyle were observed in three different cell lines (HeLa, HL-60 and hESC) with diverse nuclear morphology. H3K56ac is concentrated to several hundreds focuses in a cell nuclei. We observe the distribution of H3K56ac focuses in a cell nuclei. Amount of H3K56ac focuses increases during cell cycle progression in S and G2 phase in malignant cell lines, whereas it remains equal in ESC. Although the level of acetyted protein itself in malignant cell lines increases, the H3K56ac:H3 ratio is constant during cell cycle progression. Thus, H3 enhanced level in malignant cell lines is accompanied by increased level of H3K56ac. Our results show H3K56ac dynamic distibution during cell cycle.
