Methods in genetic association studies

Abstract:

V bakalářské práci se věnuji hodnocení genetických asociačních studií. Genetické asociační studie hledají asociaci mezi genetickými markery a výskytem onemocnění na základě rozdílností jedinců ve studované populaci. Práce se konkrétně zaměřuje na hodnocení asociace jednonukleotidových polymorfizmů a stavu jedince: pacient/kontrola. Tyto studie jsou schopny odhalit i malé vlivy jednotlivých úseků DNA, které jsou častou příčinou dosud nerozluštěných komplexních onemocnění. Bakalářská práce začíná popisem struktury DNA a pokračuje přehledem metod, které testují významnost asociace. Práce charakterizuje strukturu genetické informace a zdůrazňuje problematiku komplexních onemocnění. Popisuje statistické metody, které detekují vliv polymorfizmů na základě jejich marginálních vlivů nebo vlivů interakce. Práce poskytuje přehled metod pro studium modelů dědičnosti genetických variant. A naopak přehled metod, keré se jakýmkoli předpokladům vyhýbají – robustní procedury. Při studiu komplexních onemocnění hraje klíčovou roli interakce mezi studovanými markery. Práce je zakončena popisem neparametrických metod hodnocení interakcí mezi polymorfizmy. Vybrané metody jsem aplikovala na reálný datový soubor, ve kterém jsem hodnotila asociaci mezi vybranými SNP a výskytem parazita u Murgeských koní.

Abstract:

In this thesis I study the evaluation of genetic association studies. Genetic association studies look for association between genetic markers and the occurence of disease based on diversity of subjects in the study population.The work specifically focuses on the evaluation of singe nucleotide polymorphisms and association status of individuals: patient/control. These studies are able to detect even small efects of individual sections of DNA that are a common cause of complex diseases. The thesis begins with description of the structure of DNA and continues with an overwiev of methods, which test the significance of the association. The work describes the structure of genetic information and highlights the issue of complex diseases. It describes statistical methods detecting the influence of polymorphisms on the basis of their marginal effects or interaction effects. The work provides an overview of methods studying the patterns of inheritance of genetic variants. Conversely is given a survey of methods avoiding any assumptions – robust procedures. The interaction between markers plays a key role in studies of complex diseases. The work is is finished with a description of nonparametric methods to assess the interaction between polymorphisms. I applied selected methods to real data set in which I evaluated the association between selected SNPs and the occurrence of the parasite in the Murgese horses.
